摘要
试验旨在研究蓝刺头多糖提取物对试验小鼠抗氧化性能、免疫机能的影响及其急性毒性作用,为后续蓝刺头相关饲料添加剂的研究与开发提供试验依据。试验选用体重相近的昆明种小鼠随机分为五组,分别为空白对照组、阳性对照组和蓝刺头多糖高、中、低剂量组。试验进行21 d后,试验小鼠眼球采血,脱颈处死后分离并称量脾脏、肝脏和胸腺等器官重量,分离血清用于血清IgG、LZM以及TNF-α含量的测定,肝脏研磨匀浆之后用于检测肝脏SOD、MDA、CAT、GSH-Px等抗氧化酶活性,与此同时通过急性经口毒性试验、骨髓细胞微核试验和精子畸形试验初步判定蓝刺头多糖提取物对昆明小鼠的毒性作用。结果显示:蓝刺头多糖提取物可显著提高试验小鼠免疫器官指数、血清IgG含量、TNF-α含量和LZM含量(P<0.05),具有提高小鼠肝脏CAT活性的趋势。且灌喂蓝刺头多糖提取物的试验小鼠在整个试验期内无异常行为表现及死亡,小鼠骨髓嗜多染红细胞微核试验结果和小鼠精子畸形试验结果显示蓝刺头多糖提取物对试验小鼠无致畸作用。由此证明蓝刺头多糖提取物对试验小鼠无毒副作用,且可提高试验小鼠抗氧化机能与免疫功能。
The purpose of this experiment was to study the effects of Echinops latifolius Tausch poly⁃saccharides's immune function,antioxidant function and acute toxicity of mice,and provide experimen⁃tal basis for subsequent research and development of related feed additives.The Kunming mice of simi⁃lar weight were randomly divided into five groups,namely blank control group,positive control group and Echinops latifolius Tausch polysaccharides's high,medium and low dose groups.After 21 days,the blood was collected from the eyeballs of the mice,and the organs such as spleen,liver,and thy⁃mus were isolated and weighed after being decapitated and sacrificed.The separated serum was used for the determination of serum IgG,LZM and TNF-α.The liver was used for detect liver SOD,MDA,CAT,GSH-Px activities.The acute oral toxicity test,bone marrow cell micronucleus test and sperm abnormality test were used to preliminarily deter⁃mine the toxic effect of the Echinops latifolius Tausch polysaccharide extract on Kunming mice.The results showed that the immune organ index,serum IgG content,TNF-αcontent and LZM content were increased significantly in mice(P<0.05),had a tendency to increase the activity of CAT in the liver of mice.The toxicity ofEchinops latifolius Tausch polysaccharides on Kunming mice was determined by acute oral toxicity test,bone marrow cell micronucleus test and sperm abnormality test.The results showed that,after administration,the test mice showed no abnormal behavior,and there was no death during the whole gastric perfusion period.The mouse bone marrow polychromatic erythrocyte micronucleus test results and the mouse sperm abnormality test results showed the Echi⁃nops latifolius Tausch polysaccharides has no teratogenic effect on experimental mice.It is proved that Echinops latifolius Tausch polysaccharides has no toxic effects on the test mice,and has immunomodu⁃latory activity and antioxidant activity.
