摘要
通过16S rRNA高通量测序技术研究抗生素对SD大鼠肠道菌群多样性和结构的影响。SD大鼠40只随机分为4组:空白对照组(M0)10只、抗生素1周(M1)10只、抗生素2周(M2)10只,抗生素3周(M3)10只。其中空白组使用生理盐水灌胃,1 mL/次,2次/d,抗生素组用0.031 g/mL的氨苄西林钠,1 mL/次,2次/d灌胃,分别在1d、7d、14d、21d处死M0、M1、M2、M3组的10只SD大鼠,收集大鼠肠道内容物。提取样本DNA进行16S rRNA高通量测序并分析。结果表明,抗生素不同用药时长与空白对照组的肠道菌群的结构存在明显差异。与M0组相比,M1组的大鼠肠道菌群摩根氏菌属(Morganella)的丰度上升,苏黎世杆菌属(Turicibacter)丰度下降;M2组的大鼠肠道毛螺菌属(Lachnospiracea_incertae_sedis)丰度上升,Terrisporobacter丰度下降;M3组差异最小;抗生素能引起尿素循环降低和甲酰基-四氢叶酸的生物合成途径的升高。研究结果表明,广谱类抗生素氨苄西林钠破坏肠道菌群的稳态,能增加结肠息肉和抑郁症等疾病的风险,但在白血病和帕金森疾病上有潜在治疗价值。
To study the effect of antibiotics on the diversity and structure of intestinal microbiota of SD rats by 16S rRNA high-throughput sequencing technology,forty SD rats were randomly divided into 4 groups:ten in blank control group(M0),10 in the group applying antibiotics for 1 week(M1),10 in the group applying antibiotics for 2 weeks(M2),and 10 in the gruop applying antibiotics for 3 weeks(M3).The blank group was administered with normal saline,1 mL/time,2 times/d,and the antibiotic group was administered 0.031 g/mL of ampicillin sodium,1 ml/time,2 times/d,respectively.At 1 d,7 d,14 d,and day 21,10 SD rats in the M0,M1,M2,and M3 groups were sacrificed,and the intestinal contents of the rats were collected.Sample DNA was extracted for 16S rRNA high-throughput sequencing and analysis.Results showed there were significant differences in the structure of intestinal flora between different antibiotics and the control group.Compared with the M0 group,the abundance of Morganella in the intestinal flora of the M1 group increased,and the abundance of the Turicibacter decreased.In the M2 group,the abundance of Lachnospiracea_incertae_sedis increased,and the Terrisporobacter abundance decreased.The M3 group had the smallest difference.Antibiotics caused a decrease in urea cycle and an increase in the formyl-tetrahydrofolate biosynthetic pathway.Thus it is concluded that the broad-spectrum antibiotic ampicillin sodium disrupts the homeostasis of the intestinal flora and can increase the risk of diseases such as colon polyps and depression,but it has potential therapeutic value in leukemia and Parkinson's disease.
