摘要
为探索紫苏叶花色苷的绿色提取技术,文章研究了聚乙二醇提取紫苏叶花色苷工艺。在单因素试验的基础上,选择聚乙二醇浓度、液料比、提取温度、提取时间为影响因素,花色苷得率为响应值,采用Box-Benhnken设计(BBD)优化提取工艺;同时,对优化工艺提取的花色苷进行抗氧化性能研究。结果表明:优化的聚乙二醇紫苏叶花色苷提取工艺条件为聚乙二醇浓度76%,液料比22.5:1 mL/g,提取温度53℃,提取时间63 min;在此条件下,紫苏叶花色苷得率为6.627 mg CGE/g DW,接近预测值(6.671 mg CGE/g DW)。紫苏叶花色苷具有良好的抗氧化能力,均表现出一定质量浓度依赖性,在100μg/mL质量浓度时对羟自由基(·OH)、超氧阴离子自由基(O2—·)、1,1-二苯基-2-三硝基苯肼自由基(DPPH·)的清除率分别为67.56%、61.82%和71.21%,在此浓度下紫苏叶花色苷还原力为Vc的57.74%。结果表明,运用响应面法优化聚乙二醇提取紫苏叶花色苷工艺具有良好效果,同时提取物具有较强的抗氧化活性。
Perilla leaves were used as material to extract anthocyanins by polyethylene glycol.On the basis of single factor experiments,the concentration of polyethylene glycol,the ratio of liquid to material,the extraction temperature and the extraction time were selected as the affecting factors.The yield of anthocyanins was the response value.The extraction process was optimized by Box-Benhnken design(BBD).Meanwhile,the antioxidant activity of anthocyanins from perilla leaves under the optimized conditions was studied.The results showed that the optimum extraction conditions of anthocyanins from perilla leaves were as follows:the concentration of polyethylene glycol was 76%,the ratio of liquid to solid was 22.5:1 mL/g,the extraction temperature was 53℃,and the extraction time was 63 min.Under these conditions,the yield of anthocyanins from perilla leaves was 6.627 mg CGE/g DW,while the predicted value was 6.671 mg CGE/g DW.Anthocyanins from perilla leaves exhibited good antioxidant activity in a concentration-dependent manner.At the concentration of 100μg/mL,the scavenging rates of anthocyanins from perilla leaves on hydroxyl radicals(·OH),superoxide anion radicals(O2—·),1,1-diphenyl-2-trinitrophenylhydrazine radicals(DPPH·)were 67.56%,61.82%and 71.21%,respectively.Meanwhile,the reducing power of anthocyanins from perilla leaves was 57.74%of Vc.The results showed that the response surface methodology was effective in optimizing the extraction process of anthocyanins from perilla leaves by polyethylene plycol,and the extract had strong antioxidant activity.
