祛积通络方对RVO大鼠视网膜HIF-1α、VEGF mRNA表达的影响 被引量:8

摘要 目的探讨祛积通络方对视网膜静脉阻塞(RVO)大鼠视网膜HIF-1α、VEGF mRNA表达的影响。方法60只棕色挪威(BN)大鼠随机分为正常空白组(正常组),模型组,阳性对照组(对照组),祛积通络低、中、高剂量组(低、中、高剂量组),共6组,每组10只。除正常组不做处理、正常饲养外,其他各组大鼠右眼均用光化学动力法建立RVO动物模型。造模成功后,模型组大鼠予蒸馏水灌胃,对照组大鼠予复方血栓通胶囊灌胃,低、中、高剂量组予不同剂量祛积通络方灌胃。分别于造模后1 h、28 d,利用RT-PCR检测大鼠视网膜组织中HIF-1α、VEGF mRNA的表达。结果(1)HIF-1αmRNA:造模后1 h,各组间比较,差异无统计学意义(F=2.489,P=0.088)。造模后28 d,对照组和低、中、高剂量组表达水平相对于模型组均下降,组间比较,差异有统计学意义(F=7.981,P=0.007);中、高剂量组与对照组比较,t中剂量组=3.071,P=0.037;t高剂量组=4.620,P=0.010;中、高剂量组之间比较,t=3.147,P=0.035,差异均有统计学意义。(2)VEGF mRNA:造模后1 h,各组间比较,差异无统计学意义(F=4.327,P=0.055)。造模后28 d,对照组和低、中、高剂量组表达水平相对于模型组均下降,组间比较,差异有统计学意义(F=5.563,P=0.021);对照组与三种剂量组比较,t低剂量组=3.221,P=0.032;t中剂量组=4.349,P=0.012;t高剂量组=5.689,P=0.005;低剂量组与中、高剂量组比较,t中剂量组=3.113,P=0.032;t高剂量组=7.612,P=0.001;中、高剂量组比较,t=5.805,P=0.005,差异均有统计学意义。结论光化学动力法诱导的大鼠RVO模型视网膜HIF-1α、VEGF mRNA的表达水平上调,祛积通络方对视网膜HIF-1α、VEGF mRNA的表达有抑制作用。 OBJECTIVE To investigate the effect of Quji Tongluo decoction on the expression of HIF-1αand VEGF mRNA in retina of rats with retinal vein occlusion(RVO).METHODS A total of 60 Brown Norway(BN)rats were randomly divided into the normal group,the model group,the control group(CG),low dose of Quji Tongluo decoction group(LG),medium dose of Quji Tongluo decoction group(MG),and high dose of Quji Tongluo decoction group(HG)evenly.Except for the normal group,which was left untreated and fed normally,rats of other groups were established as RVO animal model by photochemical dynamic method in the right eye.After successful modeling,rats in the model group were treated with distilled water by gavage,rats in the control group were treated with compound Xueshuantong capsule by gavage,and rats in the low,medium,and high group were treated with different doses of Quji Tongluo decoction by gavage.After 1 hour and 28 days of administration,the expression of HIF-1αand VEGF mRNA in rat retina was detected by RT-PCR.RESULTS(1)HIF-1αmRNA:One hour after treatment,there was no significant difference in retinal expression among the groups(F=2.489,P=0.088).Twenty-eight days after treatment,the expression levels of the control group,low,medium and high dose group were lower than that of the model group,and the difference between the groups was statistically significant(F=7.981,P=0.007).The medium and high dose groups were compared with the control group(tMG=3.071,P=0.037;tHG=4.620,P=0.010);result of medium and high dose groups was compared(t=3.147,P=0.035),the differences were all statistically significant.(2)VEGF mRNA:One hour after treatment,there was no significant difference in retinal expression among the groups(F=4.327,P=0.055).Twenty-eight days after treatment,the expression levels of the control group,low,medium and high dose group were lower than that of the model group,and the difference between the groups was statistically significant(F=5.563,P=0.021).The control group was compared with the different dose groups(tLG=3.221,P=0.032;tMG=4.349,P=0.012;tHG=5.689,P=0.005);the low dose group was compared with the medium and high dose groups(tMG=3.113,P=0.032;tHG=7.612,P=0.001);the medium dose group was compared with the high dose group(t=5.805,P=0.005),the differences were statistically significant.CONCLUSIONS The expression levels of HIF-1αand VEGF mRNA in retinal of rat RVO model induced by photochemical dynamic method were up-regulated,and Quji Tongluo decoction could inhibit the expression of HIF-1 α and VEGF mRNA in retina.
