摘要
目的探究柴胡皂苷D抑制线粒体凋亡通路对心肌缺血再灌注大鼠心脏功能影响及作用机制.方法75只大鼠分为Control组、MI/R组、低、中、高剂量柴胡皂苷D组,15只/组;造模后,用药组分别使用1、3、6 mg/kg的柴胡皂苷D处理;检测心脏功能、血清心损标志物、炎性因子、氧化应激标志物、凋亡蛋白、心肌凋亡水平;观察各组大鼠心肌组织结构变化.结果与Control组比较,MI/R组HR、MAP、LVEF、FS、SOD、GSH和c-Myc蛋白水平降低(P<0.05);Mb、CK-Mb、cTnI、MDA、心肌细胞凋亡率、血清TNF-α、心肌组织TNF-α、Bax/Bcl-2、cleavCaspsae-3/Caspase-3、cleavCaspsae-9/Caspase-9水平升高(P<0.05);血清IL-10、心肌组织IL-10水平无显著性差异(P>0.05);相比MI/R组,中、高剂量柴胡皂苷D组上述指标变化显著改善(P<0.05).结论柴胡皂苷D可通过抑制炎性反应防止线粒体凋亡实现保护缺血在灌注大鼠心肌组织.
Objective To explore the ffcts and action mechanism of saikosaponin-D(SAID)on cardiac function of rats undergoing myocardial ischemia-reperfusion(MI/R)by inhibiting mitochondrial apoptosis pathway.Methods Seventy-five rats were divided into control group,MI/R group,low-dose,medium-dose and high-dose SAI-D groups,15 cases in each group.After the establishment of the model,rats in administration groups were treated with 1,3 and 6 mg/kg SAI-D,respectively.The cardiac function,levels of serum myocardial injury markers,inflammatory factors,oxidative stress markers,apoptosis proteins and myocardial apoptosis were detected.The changes of myocardial tissue structure in each group were observed.Results The levels of HR,MAP,LVEF,FS,SOD,GSH and c-Myc protein were significantly decreased(P<0.05),while the levels of Mb,CK-Mb,cTnI,MDA,cardiomyocyte apoptosis rate,serum TNF-α,TNF-α,Bax/Bcl-2,cleaved Caspsae-3/Caspase-3 and cleaved Caspsae-9/Caspase-9 in myocardial tissues were significantly increased in MI/R group compared with control group(P<0.05).There were no significant differences in levels of IL-10 in the serum and myocardial tissues between control group and MI/R group(P>0.05).The changes of the above indexes were significantly improved in medium-dose and high-dose SAI-D groups compared with MI/R group(P<0.05).Conclusion SAI-D can prevent mitochondrial apoptosis by inhibiting inflammatory response,so as to protect myocardial tissues of MIR rats.
