摘要
目的探讨eIF3A高表达对喉癌Hep-2细胞顺铂敏感性的影响及其可能机制。方法通过慢病毒感染的方式构建eIF3A高表达的Hep-2稳转细胞及对照Hep-2细胞,两组细胞传代至对数生长期后均利用顺铂进行处理,处理后的24及48h分别计算各组细胞的生长抑制率,处理5天后通过qRT-PCR法检测两组细胞Caspase3mRNA及Survivin mRNA分别的表达情况,最后对各组数据进行统计分析。结果细胞培养24h后,处理组及对照组细胞生长抑制率分别为25.6±5.8及4.1±0.3,差异具有统计学意义(P<0.001);细胞培养48h后,处理组及对照组细胞生长抑制率分别为51.4±7.5及4.0±0.4,差异具有统计学意义(P<0.001)。细胞培养5天后,处理组及对照组细胞Caspase3mRNA表达水平分别为1.93±0.27及0.84±0.14,差异具有统计学意义(P<0.001);处理组及对照组细胞Survivin mRNA表达水平分别为0.52±0.14及1.34±0.17,差异具有统计学意义(P<0.001)。结论eIF3A高表达能够增加喉癌Hep-2细胞对顺铂的敏感性,调节Caspase3及Survivin的表达。
Objective To investigate the effect of eIF3 aoverexpression on cisplatin sensitivity of HEp-2 cells and its possible mechanism.Methods eIF3 ahighly expressed Hep-2 stably transformed cells and control HEp-2 cells were constructed by lentivirus infection.The cells in the two groups were treated with cisplatin after passage to the logarithmic growth period.The growth inhibition rates of each group were calculated at 24 and 48 hours after treatment.Caspase-3 mRNA and Survivin were detected by qRT-PCR at 5 days after treatment.The expression of mRNA was analyzed.Results After 24 hours of cell culture,the cell growth inhibition rates of the treatment group and the control group were 25.6±5.8 and 4.1±0.3 respectively,the difference was statistically significant(P<0.001);after 48 hours of cell culture,the cell growth inhibition rates of the treatment group and the control group were 51.4±7.5 and 4.0±0.4 respectively,the difference was statistically significant(P<0.001).After 5 days of cell culture,the expression level of caspase 3 mRNA in the treatment group and the control group was 1.93±0.27 and 0.84±0.14,respectively,with statistical significance(P<0.001);the expression level of survivin mRNA in the treatment group and the control group was 0.52±0.14 and 1.34±0.17,respectively,with statistical significance(P<0.001).Conclusion The high expression of eIF3 acan increase the sensitivity of HEp-2 cells to cisplatin,it can regulate the expression of caspase 3 and survivin.
