摘要
目的探讨补阳还五汤对脑缺血后大鼠神经功能的影响。方法采用改良线栓法制备大鼠大脑中动脉栓塞模型,补阳还五汤灌胃后以Longa 5级4分法评价模型的神经功能缺失症状。神经功能评分后断头取出鼠脑,采用实时荧光定量PCR扩增LAMP2基因。结果术后第7天,补阳还五汤灌胃治疗后大鼠神经功能得到一定程度的恢复,各组间评分和恢复程度有差异。LAMP2 mRNA正常组、低剂量组、中剂量组、高剂量组mRNA相对表达水平较模型组均上调(均P<0.01)。正常组表达量最高,高剂量组高于中剂量组,中剂量组高于低剂量组,低剂量组大于模型组(均P<0.01)。结论补阳还五汤能通过上调LAMP2表达影响CMA的活性,进而启动自噬调控机制改善脑缺血后大鼠的神经功能。
Objective To discusion the effect of Buyang Huanwu decoction on nerve function of rats after cerebral ischemia.Methods The model of middle cerebral artery embolism in rats was made by the improved thread embolism method,and the neurological symptoms of the model were evaluated by the longa 5-grade 4-point method after the Buyang Huanwu decoction was infused into the stomach.After the neurological score,the rat brain was decapitated and the Lamp2 gene was amplified by real-time fluorescence quantitative PCR.Results On the 7th day after operation,the nerve function of rats was recovered to a certain extent after the treatment of Buyang Huanwu decoction by gavage,and there were differences in scores and recovery degrees among groups.Compared with the model group,the relative mRNA expression level of Lamp2 normal group,low dose group,middle dose group and high dose group were all up-regulated(P<0.01).The expression level of normal group was the highest,high dose group was higher than middle dose group,middle dose group was higher than low dose group,low dose group was higher than model group(P<0.01).Conclusion Buyang Huanwu decoction can affect the activity of CMA by up regulating the expression of Lamp2,and then start the regulatory mechanism of autophagy to improve the neural function of rats after cerebral ischemia.
