摘要
目的采用一测多评法同时测定骨疏丹中12种有效成分(淫羊藿苷、朝藿定A、朝藿定B、朝藿定C、淫羊藿次苷Ⅱ、蛇床子素、欧前胡素、佛手内酯、新北美圣草苷、柚皮苷、丹酚酸B、丹参酮ⅡA)。方法色谱柱为Cosmosil 5C18-MS-Ⅱ柱(250 mm×4.6 mm,5μm);流动相为乙腈-体积分数0.1%甲酸水系统,梯度洗脱,流速为1.0 mL·min^-1,检测波长为272 nm和285 nm。结果方法学结果表明各待测成分线性关系良好(r≥0.999 0),各待测成分的回收率在95.6%~106.8%之间(RSD<3.9%)。以淫羊藿苷为内参比物质,各待测成分校正因子的重现性良好(RSD<5%)。斜率校正因子法、多点平均校正因子法和标准曲线法计算的结果间没有显著性差异(RSD<3.7%,P>0.1)。结论建立的一测多评法简便、快速、准确,可用于骨疏丹的质量评价研究。
Objective To develop a quantitative analysis of multi-components with single-marker method(QAMS) for simultaneous determination of 12 active constituents:Neoeriocitrin,Naringin,Salvianolic acid B,Epimedin A,Epimedin B,Epimedin C,Icariin,Bergapten,Icariside Ⅱ,Imperatorin,Osthole,Tanshinone ⅡA in Gushudan decoction and to calculate two kinds of relative correction factors the linear regression-relative correction factor(LRG-RCF) and the average-relative correction factor(AVG-RCF).Methods The analysis was carried out on a HPLC-DAD with a column of Cosmosil 5 C18-MS-Ⅱ(250 mm×4.6 mm,5 μm).The mobile phase was water with 0.1% formic acid(A) and acetonitrile(B) with a gradient elution at a flow rate of 1.0 mL·min^-1 and the multi-wavelength was 272 nm and 285 nm.Results The result showed that all the twelve constituents had good linearity(r≥0.999 0).The recovery was ranged from 95.6% to 106.8% and the RSD were lower than 3.9%.Icariin was selected as internal reference substances and the relative correction factors(RCF)of other 11 components RCFs were consistent at different instruments,chromatographic columns,column temperatures and flow rates with the RSD were lower than 5%.The result showed that there were no significant differences between external reference method and QAMS.Conclusion Simultaneous determination of 12 active constituents in Gushudan by QAMS can be used for the analysis of quality control research of Gushudan decoction.
