基于超高效液相色谱-高分辨质谱联用的非靶向代谢组学分析普洱生茶和普洱熟茶醇溶性物质组成差异 被引量:6

摘要 [目的]建立了基于超高效液相色谱-高分辨质谱联用的非靶向代谢组学方法分析普洱生茶和普洱熟茶中醇溶性差异物质。[方法]通过70%甲醇提取和液相色谱-高分辨质谱分析,结合保留时间的规律和数据库匹配鉴定物质,依据正交偏最小二乘法-判别分析表明两者间物质存在显著差异,结合模型的变量重要因子和非参数检验,筛选差异物质。[结果]普洱生茶和普洱熟茶中共鉴定出100个化合物。通过统计分析,2种普洱茶中筛选出32个差异物质,其中氨基酸类、嘌呤类、儿茶素类和没食子儿茶素类等物质在2组中的含量差异显著。[结论]建立的方法为合理科学解释普洱生茶和普洱熟茶功效提供一定的依据。 [Objective]To discover the methanol-soluble differential chemical compound in raw Pu-er tea and ripe Pu-er tea by a non-targeted metabolomics method based on UPLC-HRMS.[Method]After the liquid extraction,Ultra High Performance Liquid Chromatography-High Resolution Mass Spectrometry(UPLC-HRMS)was used for the data acquisition.The chemical identification was carried out by comparing with the known compounds in commercial and public databases.Multivariate analysis,Mann-Whitneg U test and orthogonal projections to latent structures-discrimination analysis(OPLS-DA)analysis were used to find and visualize differential chemical compound.[Result]100 compounds were tentatively identified in raw Pu-er tea and ripe Pu-er tea.Finally,32 compounds,among which,amino acids,purines,catechins,and epigallocins,were a significant difference between them.[Conclusion]This study demonstrates the potentials and power of metabolomics method to understand the chemical secrets of Pu-er tea,which could provide a scientific explanation for the benefit of different Pu-er tea.
