摘要
目的:观察白芍不同炮制品对特发性血小板减少性紫癜(ITP)的作用。方法:取BALB/c小鼠分为正常对照组、模型对照组、生白芍有效组分组、麸炒白芍有效组分组。正常对照组不做处理,其余各组于第1、3、5、7、9天按每次每只0.1mL的剂量腹腔注射稀释4倍的血小板血清(APS),造模第1天开始各组给予相应溶剂和药物,每日1次,连续给药9d,造模第11天处死动物,取血和脏器进行相关指标检测。结果:经生白芍、麸炒白芍有效组分干预后,ITP小鼠血小板计数和血小板压积显著升高(P<0.01),平均血小板体积和血小板分布宽度显著降低(P<0.01),且麸炒白芍抑制ITP小鼠血小板减少的作用较强;经生白芍有效组分干预后,血清干扰素-γ(IFN-γ)水平显著升高(P<0.05),白介素-4(IL-4)水平略微降低,经麸炒白芍有效组分干预后,血清IFN-γ水平显著升高(P<0.01),IL-4水平显著降低(P<0.05);经生白芍、麸炒白芍有效组分干预后,脾脏中调节性T细胞(Treg)比例、Foxp3 mRNA表达水平均显著升高(P<0.05),且麸炒白芍升高ITP小鼠Treg比例和Foxp3 mRNA表达水平的作用较强。结论:白芍不同炮制品均能通过调节血小板计数、Th1/Th2平衡及Treg比例,从而发挥对ITP的治疗作用,且麸炒白芍对ITP的治疗效果较优。
Objective:To observe the effects of different processed of Paeoniae Radix Alba on idiopathic thrombocytopenic purpura.Methods:BALB/c mice were divided into normal control group,model control group,effective group of Paeoniae Radix Alba and effective group of bran fried Paeoniae Radix Alba.The normal control group was not treated.The other groups were intraperitoneally injected with 4 times diluted platelet serum(APS)on the 1 st,3 rd,5 th,7 th,and 9 th day at the dose of 0.1 m L per day.The corresponding solvent and drug were administered once a day for 9 consecutive days,and on the 11 th day,the animals were sacrificed,blood and viscera were taken for related indicators.Results:After the intervention of effective components of Paeoniae Radix Alba and bran fried Paeoniae Radix Alba,the number of platelets and platelet volume in mice were significantly increased(P<0.01),the average platelet volume and the width of platelet distribution were significantly decreased(P<0.01),and the effects of bran fried Paeoniae Radix Alba on inhibition of thrombocytopenia is better than Paeoniae Radix Alba.After the intervention of effective components of Paeoniae Radix Alba,serum IFN-γlevel was significantly increased(P<0.05),and IL-4 level was slightly decreased.After the intervention of effective components of bran fried Paeoniae Radix Alba,serum IFN-γlevel was significantly increased(P<0.01),and IL-4 level was significantly decreased(P<0.05).After the intervention of effective components of Paeoniae Radix Alba and bran fried Paeoniae Radix Alba,the proportion of Treg and the expression level of Foxp3 mRNA in spleen were significantly increased(P<0.05),and the effects of bran fried Paeoniae Radix Alba on the proportion of Treg and the expression level of Foxp3 mRNA is better than Paeoniae Radix Alba.Conclusion:All processed products of Paeoniae Radix Alba can regulate the number of platelets,Th1/Th2 balance and proportion of Treg,so as to play a role in the treatment of idiopathic thrombocytopenic purpura,and the effects of bran fried Paeoniae Radix Alba on idiopathic thrombocytopenic purpura is better than Paeoniae Radix Alba.
