摘要
目的:筛选抗纤益心方干预扩张型心肌病(DCM)大鼠的差异表达蛋白质,分析探讨抗纤益心方治疗DCM的分子作用机制。方法:雄性Wistar大鼠随机分为3组:正常对照(control)组、模型(model)组、抗纤益心方(KXYX)组,其中control组正常饮水饮食,不施加任何干预,model组以同体积0.9%氯化钠溶液灌胃,KXYX组以抗纤益心方灌胃,连续灌胃干预4周。通过数据非依赖性的扫描模式(DIA)蛋白组学鉴定各组间差异蛋白。结果:KXYX组与model组比较,共发现52个差异蛋白,35个上调蛋白,17个下调蛋白;差异蛋白主要分布于线粒体及细胞质内;生物过程主要包括硫化物代谢过程、细胞酰胺代谢过程、内质网介导高尔基体囊泡转运、细胞氨基酸代谢过程、小分子生物合成等;主要分子功能包括催化活性、细胞骨架蛋白结合、辅酶结合、胶原蛋白结合、NAD结合、肌动蛋白结合、氧化还原酶活性等;主要富集通路包括内质网蛋白质加工、心肌收缩、氨基酸的生物合成、产热通路等。结论:KXYX组与model组的蛋白质表达存在明显差异,抗纤益心方治疗DCM的作用机制可能与修复调节差异蛋白及信号通路相关。
Objective:To screen the differentially expressed proteins of Kangxian Yixin Formula in rats with dilated cardiomyopathy(DCM),and to discuss the molecular mechanism of Kangxian Yixin Formula in the treatment of dilated cardiomyopathy.Methods:Male Wistar rats were randomly divided into three groups:normal control group(control),DCM model group(model)and Kangxian Yixin Formula group(KXYX).In the control group,normal drinking water and diet were given without any intervention.Model was given intragastric administration of the same volume of normal saline and KXYX was given intragastric administration of Kangxian Yixin Formula for 4 weeks.Differential albumen was identified by data independent scanning mode(DIA)proteomics.Results:Compared with model,52 differential proteins were found in KXYX,35 up-regulated proteins and 17 down-regulated proteins,the differential proteins were mainly distributed in mitochondria and cytoplasm,and the biological processes mainly included sulfur metabolism,cellular amide metabolism,Golgi vesicle transport mediated by endoplasmic reticulum,cellular amino acid metabolism,small molecule biosynthesis,and so on.The main MF annotation information includes catalytic activity,cytoskeleton protein binding,coenzyme binding,collagen binding,NAD binding,actin binding,redox enzyme activity,etc..The main enrichment pathway includes endoplasmic reticulum protein processing,myocardial contraction,amino acid biosynthesis,thermogenic pathway,and so on.Conclusion:There is a significant difference in protein expression between KXYX and model,and the mechanism of Kangxian Yixin Decoction in treating DCM may be related to repair and regulation of differential proteins and signal pathways..
