摘要
目的旨在研究PKM2表达与肺鳞状细胞癌患者预后的关系,探讨沉默PKM2表达对肺鳞状细胞癌生物学行为的影响及机制。方法采用免疫组织化学方法检测95例肺鳞状细胞癌与正常肺组织中PKM2与Bim蛋白表达情况,分析PKM2表达与肺鳞状细胞癌患者临床病理因素、患者生存预后的关系。采用脂质体介导法将siRNA片段导入肺鳞状细胞癌中来沉默PKM2表达。分别采用Western blot法、RT-PCR方法检测PKM2与Bim蛋白与mRNA表达。利用Annexin V-FITC/PI试剂盒检测细胞凋亡。结果PKM2在74例肺鳞状细胞癌组织中高表达(77.9%),PKM2高表达与患者高TNM分期(P=0.008)、淋巴结转移(P=0.003)及肿瘤直径(P=0.009)相关,同时PKM2高表达与肺鳞状细胞癌患者不良预后相关(P=0.000,χ^2=18.630)。SK-MES-1细胞中共转染PKM2 siRNA与Bim siRNA后,与单独转染PKM2 siRNA比较,Bim表达增加(P<0.05),细胞凋亡减少(分别为13.55%±1.74%与28.01%±3.65%,P<0.05)。结论PKM2高表达与肺鳞状细胞癌患者不良预后相关,且PKM2可通过Bim来调控肺鳞状细胞癌凋亡。
Objective To study the relationship between the expression of PKM2 and the prognosis of patients with lung squamous cell carcinoma,and to investigate the effect and mechanism of silencing the expression of PKM2 on the biological behavior of lung squamous cell carcinoma.MethodsImmunohistochemistry was used to detect the expression of PKM2 and Bim protein in 95 cases of lung squamous cell carcinoma and normal lung tissues.The correlation between the expression of PKM2 and clinicopathological factors and survival prognosis of patients with lung squamous cell carcinoma were analyzed by statistical methods.Liposome-mediated siRNA fragments were introduced into lung squamous cell carcinoma to silence the expression of PKM2.The expressions of PKM2,Bim protein and mRNA were detected by Western blot and RT-PCR.Annexin V-FITC/PI kit was used to detect cell apoptosis.ResultsPKM2 was highly expressed in 74 cases of lung squamous cell carcinoma(77.9%).The high expression of PKM2 was associated with higher TNM stage(P=0.008),lymph node metastasis(P=0.003)and tumor size(P=0.009).Meanwhile,high expression of PKM2 was associated with poor prognosis in patients with lung squamous cell carcinoma(P=0.000,χ^2=18.630).After co-transfection of PKM2 siRNA and Bim siRNA with SK-MES-1 cells,compared with transfection of PKM2 siRNA alone,the level of Bim increased(P<0.05),cell apoptosis decreased(13.55%±1.74%vs 28.01%±3.65%,respectively,P<0.05).ConclusionHigh expression of PKM2 is associated with poor prognosis in patients with lung squamous cell carcinoma,and PKM2 can regulate apoptosis of lung squamous cell carcinoma by Bim.
