摘要
角蛋白是维持毛发形态特征的重要结构蛋白,包括角蛋白和角蛋白关联蛋白。角蛋白74(Keratins 74,KRT74)属于Ⅱ型内根鞘角蛋白,与毛发的生长和形态结构多样性相关。通过探究KRT74基因的结构特征、蛋白性质及其在内蒙古绒山羊不同毛被类型的表达差异,为挖掘内蒙古绒山羊毛被类型遗传标记和绒毛生长发育研究提供理论依据。利用NCBI公布的山羊KRT74序列信息进行生物信息学分析,通过实时荧光定量PCR技术(qRT-PCR)检测该基因在内蒙古绒山羊不同毛被类型皮肤组织中的表达水平。生物信息学分析发现KRT74蛋白为亲水性稳定蛋白,没有跨膜结构和信号肽;主要分布在细胞核和细胞质中,蛋白质存在4个保守结构域,是细胞骨架和核膜的原始成分,并为上皮细胞提供机械支持。qRT-PCR结果显示,长毛型绒山羊KRT74基因的表达水平显著高于短毛型绒山羊,差异倍数为1.6;相关性分析发现KRT74基因表达量与毛长性状呈显著正相关。推测KRT74基因可能是影响内蒙古绒山羊不同毛被类型的候选基因。
Keratin is an important structural protein that maintains the morphological characteristics of hair,including keratin and keratin-associated protein.Keratin 74 belongs to typeⅡepithelial keratin,which is related to hair growth and morphological diversity.The purpose of this study is to explore the structural characteristics and protein properties of KRT74 gene and its expression in different hair types of Inner Mongolia cashmere goats,it will provide a theoretical basis for the use of genetic markers of coat type and the growth and development of wool in Inner Mongolia cashmere goat.In this study,the goat KRT74 sequence information published by NCBI was used for bioinformatics analysis.The relative expression of KRT74 gene in different types of skin tissue of Inner Mongolia cashmere goat was detected by real-time fluorescence quantitative PCR(qRT-PCR).Bioinformatic analysis showed that KRT74 protein was a hydrophilic stable protein without transmembrane structure and signal peptide,which was mainly distributed in the nucleus and cytoplasm.This protein has four conservative domains,which are the original components of cytoskeleton and nuclear membrane,and provides mechanical support for epithelial cells.The results of qRT-PCR showed that the expression level of KRT74 in cashmere goats of the long hair type was significantly higher than that in cashmere goats of the short hair type,its difference was 1.6 times.Correlation analysis showed that there was a significantly positive correlation between KRT74 expression and wool length traits.It is speculated that KRT74 may be a candidate gene affecting different hair types in Inner Mongolia cashmere goat.
