摘要
【目的】拟克隆朱砂叶螨几丁质脱乙酰基酶TecCDA3并对其进行表达特征分析。【方法】采用同源基因克隆技术克隆TecCDA3,通过实时荧光定量PCR技术分析其表达特征。【结果】成功克隆朱砂叶螨几丁质脱乙酰基酶TecCDA3(GenBank No.MK779705),其开放阅读框长度为1611 bp,共编码536个氨基酸,含有3种结构域,根据同源性和发育树分析判定其属于几丁质脱乙酰基酶家族GroupⅡ。TecCDA3在成螨时期表达量最高,在幼螨时期表达量最低,呈现先下降后升高的表达趋势。【结论】克隆得到朱砂叶螨几丁质脱乙酰基酶TecCDA3并对其表达特征进行分析,丰富朱砂叶螨几丁质脱乙酰基酶基因家族。
【Objective】This study was to clone the chitin deacetylase gene of Tetranychus cinnabarinus and to analyze its expression characteristics.【Methods】TecCDA3 was cloned using homologous gene cloning technology,and the expression characteristics were analyzed by RT-qPCR.【Results】TecCDA3 was successfully cloned from T.cinnabarinus(GenBank No.MK779705),with an open reading frame of 1611 bp coding 536 amino acids.TecCDA3 contains three domains,and belongs to Chitin deacetylase family GroupⅡaccording to homology comparison and phylogenetic analysis.The highest expression level was found at the adult stage,while the lowest expression level was at the larva stage.The expression of TecCDA3 declines at first and then increases.【Conclusion】The chitin deacetylase TecCDA3 was cloned and its expression characteristics were analyzed,which enriched the chitin deacetylase gene family of T.cinnabarinus.
