转基因玉米对蛋鸡生理特性及DNA重组表达的影响

摘要 本研究旨在研究植酸酶转基因玉米对蛋鸡组织器官相对重量、血清生化指标、养分表观消化率及DNA重组的影响。试验选择160只48周龄、平均产蛋率为76%的海兰褐壳蛋鸡,随机分为2组,每组5个重复,每个重复16只。对照组日粮中使用625 g/kg正常玉米,处理组日粮中使用625 g/kg植酸酶转基因玉米。饲养试验共进行16周。结果:转基因玉米组心脏、脾脏、肺和肾脏相对重量在数值上高于对照组,而对照组肝脏相对重量在数值上高于处理组,但均无显著差异(P>0.05)。对照组与处理组对总能、干物质、粗蛋白质和钙表观消化率的影响无显著差异(P>0.05),但转基因玉米日粮组磷表观消化率较正常玉米日粮组显著提高22.39%(P<0.05)。正常玉米与转基因玉米对血清总蛋白、白蛋白、胆固醇、甘油三酯浓度及血清谷草转氨酶、谷丙转氨酶、碱性磷酸酶、谷氨酰转移酶和乳酸脱氢酶活性无显著影响(P>0.05)。正常玉米、转基因玉米、正常玉米日粮及转基因玉米日粮中玉米特异性转化酶基因DNA重组表达达到100%。转基因玉米和转基因玉米日粮中phyA2基因DNA重组表达为100%。对照组和处理组胸肌、腿肌、卵巢、输卵管和鸡蛋中家禽特异性卵清蛋白基因DNA重组表达为100%。结论:本研究在肌肉组织和生殖器官中未检测到重组的phyA2基因,养分消化率数据表明,植酸酶转基因玉米日粮组可以提高磷表观消化率,最大限度地减少日粮中磷的添加水平。 The purpose of this study was to investigate the effects of phytase transgenic maize on the relative weight,serum biochemical index,nutrient apparent digestibility and DNA recombination of layers.Methods:A total of 16048-weekold brown shell laying hens with an average egg yield of 76%were randomly divided into 2 treatment groups with 5 replicates in each group and 8 replicates in each group.The control group was fed 625 g/kg normal corn,and the treatment group was fed 625 g/kg phytase transgenic corn.The feeding experiment was carried out for 16 weeks.Results:the relative weight of heart,spleen,lung and kidney in the transgenic maize group was higher than that in the control group,while the relative weight of liver in the control group was higher than that in the treatment group,but there was no significant difference(P>0.05).There was no significant difference between the control group and the treatment group on the apparent digestibility of total energy,dry matter,crude protein and calcium(P>0.05),but the apparent digestibility of phosphorus in the transgenic corn diet group was 22.39%higher than that in the normal corn diet group(P<0.05).Normal corn and transgenic corn had no significant effect on serum total protein,albumin,cholesterol,triglyceride concentration,and serum glutamates transaminase,alanine transaminase,alkaline phosphatase,glutamyltransferase and lactate dehydrogenase activities(P>0.05).The recombinant DNA expression of maize specific invertase gene in normal corn,transgenic corn,normal corn diet and transgenic corn diet reached 100%.The recombinant DNA expression of phyA2 gene in gm corn and gm corn diet was 100%.The recombinant DNA expression of poultry-specific egg albumin gene in pectoral muscle,leg muscle,ovary,fallopian tube and egg was 100%in control group and treatment group.Conclusion:in this study,no recombinant phyA2 gene was detected in muscle tissues and reproductive organs.Nutrient digestibility data indicated that phytase transgenic corn diet group could improve the apparent digestibility of phosphorus and minimize the dietary addition level of phosphorus.
