摘要
叉头框蛋白(Forkhead box,FOX)N2(FOXN2)是FOX蛋白家族中的一员,据报道其与肿瘤的发生发展相关,在乳腺癌、肺癌等疾病中起关键作用.为利用基因工程构建低成瘤性Madin-Darby canine kidney(MDCK)细胞系,本研究探讨FOXN2不同成瘤性MDCK细胞中的差异表达,培养不同成瘤性的MDCK细胞.通过RT-qPCR来检测FOXN2在不同成瘤性MDCK细胞中的表达.结果表明FOXN2在不同成瘤性MDCK细胞中存在差异,在高成瘤性细胞株MEDK-A和MDCK-B中表达高,在低成瘤性细胞株MDCK-C中表达量低,说明该蛋白对MDCK细胞的成瘤性能起到一定的调控作用,可为进一步研究FOXN2在不同成瘤性MDCK细胞中的作用机制奠定理论基础.
Forkhead box(FOX)N2(FOXN2)is a member of the FOX protein family.It is reported to be involved in the development of tumors and plays a key role in breast cancer,lung cancer and other diseases.To use genetic engineering to construct a tumor-free Madin-Darby canine kidney(MDCK)cell line,this study was to explore the differential expression of FOXN2 in different tumorigenic MDCK cells.MDCK cells of different tumorigenicity were cultured,and the expression of FOXN2 in different tumorigenic MDCK cells was detected by RT-qPCR.The results showed that FOXN2 was different in different tumorigenic MDCK cells,with high expression in high tumorigenic cell lines MEDK-A and MDCK-B,and low expression in low tumorigenic cell lines MDCK-C,indicating that the protein plays a certain role in regulating the tumorigenicity of MDCK cells,and lays a theoretical foundation for further research on the mechanism of FOXN2 in different tumorigenic MDCK cells.
