The role of glial cell line-derived neurotrophic factor in the pathogenesis and treatment of bipolar disorder

摘要 目的:探讨胶质细胞源性神经营养因子(GDNF)水平在双相障碍(BD)发病及治疗中的作用。方法:本研究纳入70例双相障碍患者(BD组)及70名健康对照者(正常对照组)。用酶联免疫吸附法(ELISA)测定BD组治疗前后及正常对照组血清GDNF水平。采用杨氏躁狂评定量表(YMRS)评估BD组治疗前后症状水平。采用一般线性相关分析GDNF水平的变化与YMRS评分变化的关系。结果:治疗前,BD组血清GDNF水平为显著低于正常对照组,差异有统计学意义(P<0.01);治疗后血清GDNF水平上升,与治疗前相比差异有统计学意义(P<0.01)。治疗后YMRS评分低于治疗前,差异有统计学意义(P<0.01)。GDNF水平的变化与YMRS治疗4周评分变化呈负相关(r=-0.77,P<0.01)。结论:GDNF水平降低与BD发病相关,并且其水平的升高与BD症状改善相关。 Objective:To explore the characteristics of glial cell line-derived neurotrophic factor(GDNF)and its role in predicting treatment response in Chinese Han population with bipolar disorder(BD).Method:The serum GDNF levels were tested by Enzyme-Linked Immunosorbent Assay(ELISA)when Young manic rating'scale(YMRS)scores were measured at baseline and after 4 weeks of treatment.The differences in serum GDNF levels between BD patients(n=70)and healthy controls(n=70)were compared.The serum GDNF levels were also compared between pre-treatment and post-treatment.The general line regression was used to test the change between serum GDNF levels and YMRS scores.Results:The serum GDNF levels were lower in BD patients than in healthy controls(P<0.01).After the 4 weeks treatment,the serum GDNF levels were higher in post-treatment group than in pre-treatment group(P<0.01).After the 4 weeks treatment,the YMRS score was lower in post-treatment group than in pre-treatment group(P<0.01).The GDNF level was negatively correlated with YMRS score(P<0.05).Conclusion:The GDNF was demonstrated to be potential biomarker of BD,it seems that serum GDNF levels can be used to assess the baseline severity of BD and serve as a factor to predict treatment response.
