摘要
目的研究汉黄芩素对肾结石大鼠JNK信号通路的调节作用及肾损伤修复作用。方法 15只大鼠作为对照组,其余60只大鼠采用1%乙二醇联合2%氯化铵方法建立肾结石大鼠模型,造模后的大鼠随机分为模型组及汉黄芩素低、中、高剂量组,每组15只。造模后汉黄芩素低、中、高剂量组大鼠分别按照75、150、300 mg·kg^-1灌胃给予汉黄芩素,对照组及模型组大鼠灌胃给予等量生理盐水,1次·d^-1,连续给药28 d。给药结束后,检测大鼠24 h尿量,尿pH,血清Ca^2+、血清尿素氮(BUN)及肌酐(SCr)水平,尿液Ca^2+、尿草酸(Ox)、尿柠檬酸(CA)水平,肾脏超氧化物歧化酶(SOD)活性及丙二醛(MDA)水平,做肾脏组织病理学检查,检测大鼠肾脏组织JNK mRNA水平及p-JNK、JNK水平。结果与模型组相比,汉黄芩素低、中、高剂量组大鼠24 h尿量、尿pH、CA水平、肾脏SOD活性显著升高(P<0.05),尿液Ca^2+及Ox水平、血清Ca^2+、BUN及SCr水平、肾脏MDA水平均显著降低(P<0.05),肾脏组织病理学明显改善,大鼠肾脏组织JNK mRNA水平及p-JNK、JNK水平显著降低(P<0.05)。结论汉黄芩素能够调节尿液pH及离子代谢水平,改善大鼠肾功能,缓解肾结石大鼠肾损伤,其机制可能与调节JNK信号通路有关。
Objective To determine the regulatory effect of wogonin on JNK signal pathway and the repair of renal injury in rats with renal calculi.Methods Except for 15 rats in the control group,the other 60 rats were treated with 1% ethylene glycol combined with 2% ammonium chloride to establish rat models of renal calculi.The rats were randomly divided into a model group,a low,middle and high dose wogonin groups (15 rats in each group).Wogonin was intragastrically given to the rats in the low,middle and high dose groups at 75,150 and 300 mg·kg^-1,respectively.The rats in the control group and the model group were given the same amount of normal saline once daily for 28 days.After the administration,the 24 hour urine volume,urine pH,serum Ca^2+,serum urea nitrogen (BUN) and creatinine (SCr) levels,urine Ca^2+,urinary oxalate (Ox),urinary citrate (CA) levels,renal superoxide dismutase (SOD) activity and malondialdehyde (MDA) level were measured and renal histopathological examined.The levels of JNK mRNA,p-JNK,and JNK in the kidney tissue of rats were detected.Results Compared with the model group,the 24 hour urine volume,urine pH,CA levels and kidney SOD activity in the low,middle and high dose wogonin groups were significantly increased (P<0.05),while urine Ca^2+and Ox levels,serum Ca^2+,BUN,SCr and kidney MDA levels were significantly decreased.Renal histopathology was significantly improved,and the levels of JNK mRNA,p-JNK and JNK in the kidney tissue were significantly decreased (P<0.05).Conclusion Wogonin can regulate pH in the urine and ion metabolism,improve renal function and alleviate renal injury in rats with renal calculi.The mechanism may be related to the regulation of JNK signal pathway.
