鸡血藤提取物对大鼠纤维化肝细胞中胶原蛋白表达及TGF-β1/p38MAPK信号通路的影响 被引量:20

摘要 目的:探讨鸡血藤提取物对大鼠肝纤维化肝细胞中胶原蛋白表达及转化生长因子β1(TGF-β1)/p38丝裂原活化蛋白激酶(MAPK)信号通路的影响。方法:采用二甲基亚硝胺(DMN)腹腔注射法构建肝纤维化动物模型,分离纤维化肝细胞。用鸡血藤提取物处理纤维化肝细胞,MTT法检测细胞活力,Western blot检测Ⅰ型胶原蛋白(ColⅠ)、TGF-β1/p38MAPK信号通路相关蛋白(TGF-β1和p38MAPK)、细胞增殖相关蛋白(P21和CyclinD1)的表达水平。结果:与正常肝细胞相比,纤维化肝细胞ColⅠ、TGF-β1和p38MAPK蛋白的表达显著升高。鸡血藤提取物可显著抑制纤维化肝细胞的增殖活力,抑制ColⅠ、TGF-β1和p38MAPK蛋白表达。激活TGF-β1/p38MAPK信号通路能逆转鸡血藤提取物对纤维化肝细胞增殖及ColⅠ表达的影响。结论:鸡血藤提取物可通过抑制TGF-β1/p38MAPK信号通路抑制ColⅠ合成,进而发挥治疗肝纤维化的作用。 Objective:To investigate the effects of caulis spatholobi extract on collagen expression and transforming growth factorβ1(TGF-β1)/p38 mitogen-activated protein kinase(MAPK)signaling pathway in rat fibrotic hepatocytes.Methods:The rat model of liver fibrosis was established by intraperitoneal injection of dimethylnitrosamine(DMN),and the fibrotic hepatocytes were separated.Fibrotic liver cells were treated with caulis spatholobi extract,and cell activity was detected by MTT method,and the expression levels of ColⅠ,TGF-β1/p38MAPK signaling pathway related proteins(TGF-β1 and p38 MAPK)and cell proliferation related proteins(P21 and Cyclin D1)were detected by Western blot.Results:Compared with normal hepatocytes,the expressions of ColⅠ,TGF-β1 and p38MAPK in fibrotic hepatocytes were significantly increased.Caulis spatholobi extract could significantly inhibit the proliferation activity of fibrosis hepatocytes,and inhibit the expression of ColⅠ,TGF-β1 and p38MAPK.Activation of TGF-β1/p38MAPK signaling pathway reversed the effects of caulis spatholobi extract on hepatocyte proliferation and ColⅠexpression infibrotic hepatocytes.Conclusion:Caulis spatholobi extract could inhibit the synthesis of ColⅠby inhibiting TGF-β1/p38MAPK signaling pathway,and then play a role in the treatment of liver fibrosis.
