摘要
目的研究白芷多糖(ADP)对环磷酰胺所致免疫低下模型小鼠免疫功能的调节作用。方法ADP由白芷经热水煮提和乙醇醇沉提取。100只小鼠随机分为对照组、模型组和ADP低、中、高剂量(50,100,200 mg/kg)处理组,每组20只;除对照组外其余各组小鼠连续4 d腹腔注射环磷酰胺(100 mg/kg);第5~14天,ADP各处理组每天分别给予相应浓度ADP灌胃1次,对照组和模型组灌胃等体积生理盐水。计算各组小鼠免疫器官指数,通过鸡红细胞吞噬实验测定巨噬细胞吞噬率及吞噬指数,采用MTT法测定脾淋巴细胞增殖、NK细胞活性,采用酶联免疫吸附法测定血清白细胞介素2(IL-2),白细胞介素6(IL-6),肿瘤坏死因子α(TNF-α)和免疫球蛋白G(IgG)含量。结果与对照组相比,模型组小鼠的胸腺指数、脾脏指数、腹腔巨噬细胞吞噬指数和吞噬百分率、脾淋巴细胞增殖反应、NK细胞活性、血清中IL-2,IL-6,TNF-α和IgG含量均显著降低,差异具有统计学意义(均P<0.05)。与模型组比较,低剂量组小鼠TNF-α,中、高剂量组小鼠巨噬细胞吞噬指数和吞噬百分率、脾淋巴细胞增殖反应、NK细胞活性、IL-2,TNF-α,IgG以及高剂量组小鼠胸腺指数、脾脏指数、IL-6含量均显著升高,组间比较差异有统计学意义(均P<0.05)。此外,高剂量组小鼠的胸腺指数、脾脏指数、巨噬细胞吞噬指数和吞噬百分率、脾淋巴细胞增殖反应、NK细胞活性、IL-2,IL-6,TNF-α和IgG含量均显著高于低、中剂量组,组间比较差异具有统计学意义(均P<0.05)。结论ADP对环磷酰胺致免疫低下小鼠的非特异性免疫和特异性免疫具有增强作用,且该作用呈现剂量依赖性。
Objective To study the effect of Angelicae dahuricae Radix Polysaccharides(ADP)on the immune function of cyclophosphamide-induced immunosuppressive model mice.Methods ADP was extracted from Angelicae Dahuricae Radix by hot water extraction and ethanol precipitation.100 mice were randomly divided into control group,model group,and ADP low,medium,and high dose(50,100,200 mg/kg)treatment groups,20 in each group.except the normal control group,mice in other group was injected Cyclophosphamide(100 mg/kg)intraperitoneally for 4 days to establish an immunosuppressive model.On the 5 th to 14 th days,the ADP treatment groups were given intragastric administration of the corresponding concentration of ADP once a day,and the control group and the model group were given the same volume of normal saline.Calculate the immune organ index of each group of mice,determine the phagocytic rate and phagocytic index of macrophage by chicken erythrocyte phagocytosis experiment,measure the proliferation of splenic lymphocytes,NK cell activity by MTT method,and measure the serum interleukin 2(IL-2),Interleukin 6(IL-6),Tumor necrosis factor alpha(TNF-α)and immunoglobulin G(Ig G)content by ELISA.Results Compared with the control group,the thymus index,spleen index,phagocytic index and percentage of phagocytosis of peritoneal macrophages,splenic lymphocyte proliferation response,NK cell activity,the content of IL-2,IL-6,TNF-αand Ig G in serum decreased significantly,and the difference was statistically significant(all P<0.05).Compared with the model group,TNF-αin the low-dose group of mice,phagocytic index and percentage of phagocytosis of macrophages,splenic lymphocyte proliferation response,NK cell activity,IL-2,TNF-α,Ig G in the medium and high-dose group of mice,the thymus index,spleen index and IL-6 content of mice in the high-dose group were significantly increased,and the difference between the groups was statistically significant(all P<0.05).In addition,the thymus index,spleen index,phagocytic index and phagocytic percentage of macrophages,splenic lymphocyte proliferation response,NK cell activity,IL-2,IL-6,TNF-αand Ig G contents in the high-dose group were significantly higher In the low and medium dose groups,the difference between the groups was statistically significant(all P<0.05).Conclusion ADP can enhance the non-specific immunity and specific immunity of mice with immunodeficiency induced by cyclophosphamide,and the effect is dose-dependent.
