改进QuEChERS结合UPLC-MS/MS法测定梨和黄瓜中多抗霉素残留量 被引量:2

Determination of Polyoxin Residues in Pear and Cucumber by High Performance Liquid Chromatography-tandem Mass Spectrometry with Modified QuEChERS

摘要 本文建立了梨和黄瓜中多抗霉素的高效液相色谱质谱检测方法。用甲醇-水(7+3)溶液旋涡混合提取试样中多抗霉素,改进QuEChERS法净化,使用ACQUITY UPLC BEH Amide以0.1%甲酸/乙腈溶液进行梯度洗脱,采用电喷雾电离方式(ESI+),通过多反应监测(MRM)定量。结果表明,在2μg/L^50μg/L浓度范围内,多抗霉素B的线性关系良好,相关系数r均达到0.99,回收率在63%~98%之间,相对标准偏差(RSD)均小于10%。该方法快速、简便、精确度高、准确度高,对梨和黄瓜中多抗霉素的检测有普遍适用性,其他蔬菜和水果也可参考使用。 A method of the detection of polyoxin in pears and cucumbers by high performance liquid chromatography with mass spectrometry was developed. Samples ware extracted by vortex mixing with methanol-water(7+3) solution, purified with QuEChERS, applied gradient elution by ACQUITY UPLC BEH Amide with a mixture of 0.1% formic acid and acetonitrile, and finally quantified by ESI+ and MRM. The results showed that the linear relationship was good in the concentration range of 2 μg/L to 50 μg/L, and the correlation rate was more than 0.99. The average recoveries ranged from 63.0% to 98.0% with relative standard deviation less than 10%. The method is rapid, simple, sensitive and accurate. It has universal applicability to determination of polyoxin residues in pears and cucumbers and can serve as a reference for the detection of polyoxin residues in other vegetables and fruits.
