摘要
为了解甘肃沿黄灌区主栽春小麦中优质麦谷蛋白亚基的组成及分布,本研究以甘肃临夏、白银等沿黄灌区主栽的75个水地小麦品种为供试材料,采用STS分子标记的方法,选取具有代表性的优质高分子量麦谷蛋白亚基Ax1/2*、Dx5、Bx7、By8、Bx14和低分子量麦谷蛋白亚基Glu-A3d、Glu-B3b进行检测分析。结果表明,75份供试材料中,Ax1/2*、7+8和5+10分别是Glu-A1、Glu-B1和Glu-D1位点的优势亚基,分布频率分别是60.0%、54.7%、60.0%。此外,还检测到稀有亚基7、8和14+15。低分子量麦谷蛋白亚基Glu-A3d和Glu-B3b分别是Glu-A3和Glu-B3位点的优势亚基,分布频率为68.0%和56.0%。地方品种的高分子量麦谷蛋白组成单一,只有(7+8,5+10)这一种组合。3个位点中具有优质亚基的组合方式有9种,以(Ax1/2*,7+8,A3d)和(5+10,A3d,B3b)为主,各有21份材料。4个位点中具有优质亚基的组合有5种,以(5+10,7+8,A3d,B3b)为主,频率为17.3%。而5个位点中含优质亚基的组合只有1种,在6份材料中检测出,频率为8.0%。
In order to learn the composition and distribution of high quality glutenin subunits of spring wheat in irrigation districts along the Yellow River of Gansu Province,75 varieties that are mainly cultivated along the Yellow River irrigation area,such as Linxia and Baiyin in Gansu Province,were investigated by STS molecular marker in this study.As generally recognized high quality glutenin subunits,Ax1/2*,Dx5,Bx7,By8,Bx14 and Glu-A3 d,Glu-B3 b were selected for detection and analysis.The results showed that Ax1/2*,7+8 and 5+10 were the predominant subunits in their respective locus with frequency of 60.0%,54.7%,60.0%in 75 varieties,rare subunits 7,8,and 14+15 were also detected.The low molecular weight glutenin subunits,Glu-A3 d and Glu-B3 b,were the dominant subunits at The sites of Glu-A3 and Glu-B3 respectively,with a distribution frequency of 68.0%and 56.0%.The high molecular weight glutenin subunits(HMW-GS)of local varieties had a single composition consisting of 7+8 and 5+10 subunits.There were nine combinations of high-quality subunits at three sites,Ax1/2*,7+8,A3 d and 5+10,A3 d,B3 b were the dominant combinations,five combinations of high-quality subunits at four sites,5+10,7+8,A3 d,B3 b was the dominant combination with the frequency of 17.3%.There was only one combination of high-quality subunits at five sites with the frequency of 8.0%,detected in six varieties.
