摘要
目的:研究蒺藜皂苷对胃癌细胞AGS增殖和凋亡的影响及机制。方法:以胃癌细胞AGS作为研究对象,用蒺藜皂苷处理细胞,噻唑蓝(MTT)检测细胞增殖,碘化丙啶(PI)单染法检测细胞周期,膜联蛋白V-FITC(Annexin V-FITC)/PI双染法检测细胞凋亡,Western blot检测细胞周期蛋白D1(cyclin D1)、剪切的含半胱氨酸的天冬氨酸蛋白水解酶3(cleaved caspase3)、Nin1结合蛋白(NOB1)蛋白表达。在胃癌细胞中转染pc DNA3.1-NOB1,给予蒺藜皂苷处理,检测细胞增殖、周期、凋亡和Cyclin D1、cleaved caspase 3、NOB1蛋白表达变化。结果:蒺藜皂苷处理后,胃癌细胞增殖能力和Cyclin D1、NOB1蛋白表达水平下降(P<0.05),细胞G1期比例、细胞凋亡率、cleaved caspase 3蛋白表达水平升高(P<0.05)。转染pc DNA3.1-NOB1后,胃癌细胞增殖能力和Cyclin D1、NOB1蛋白表达水平升高(P<0.05),细胞G1期比例、细胞凋亡率、cleaved caspase 3蛋白表达水平下降(P<0.05)。结论:蒺藜皂苷通过下调NOB1抑制胃癌细胞增殖并诱导细胞凋亡。
Objective: To study the effect and mechanism of gross saponins of tribulus terrestris on the proliferation and apoptosis of gastric cancer cell AGS. Methods: Gastric cancer cell AGS was used as the research object,and treated with gross saponins of tribulus terrestris. MTT was used to detect the cell proliferation,PI single staining was used to detect the cell cycle,Annexin V-FITC/PI double staining was used to detect the cell apoptosis,and Western blot was used to detect the expressions of Cyclin D1,cleaved caspase 3 and NOB1 protein. pc DNA3.1-NOB1 in gastric cancer cells was transfected and treated with gross saponins of tribulus terrestris,and the cell proliferation,cycle,apoptosis and expression changes of Cyclin D1,cleaved caspase 3 and NOB1 protein were detected. Results: The proliferation,and protein expression levels of Cyclin D1 and NOB1 of gastric cancer cells after treatment with gross saponins of tribulus terrestris decreased( P<0.05),and the cell G1 phase ratio,cell apoptosis rate,and protein expression level of cleaved caspase 3 increased( P<0.05). After pc DNA3.1-NOB1 was transfected,the proliferation and protein expression levels of Cyclin D1 and NOB1 increased( P < 0.05),and the cell G1 phase ratio,cell apoptosis rate,and protein expression level of cleaved caspase 3 decreased( P< 0.05). Conclusion: Gross saponins of tribulus terrestris can inhibit the proliferation of gastric cancer cells and induce the apoptosis by down-regulating NOB1.
