摘要
目的建立对照品替代法测定中药饮片或中成药中绿原酸含量的方法。方法以对乙酰氨基酚为对照品替代,采用高效液相色谱法(HPLC),Diamonsil C18(250 mm×4.6 mm,5μm)色谱柱,流动相为乙腈-0.5%磷酸溶液,梯度洗脱,检测波长240 nm,流速1.0 ml/min,柱温30℃。利用相对斜率法测定对乙酰氨基酚与绿原酸的相对校正因子(f),计算供试品中绿原酸的含量。结果对乙酰氨基酚和绿原酸分别在5.36~107.19μg/ml(r=1.0000),5.13~102.60μg/ml(r=0.9999)浓度范围内线性关系良好;对照品替代法和外标法的平均回收率分别为98.33%,98.65%;RSD分别为1.34%,1.30%;相对校正因子f=0.5119;不同仪器及色谱柱、流速、柱温、不同测定波长对f值无显著影响;将两法的结果进行对比无显著性差异。结论以对乙酰氨基酚替代绿原酸作为对照品,采用HPLC测定绿原酸的含量进行质量控制是可行的,可用于含绿原酸成分的中药饮片及中成药的含量测定。
Objective To establish a method for the determination of chlorogenic acid in Chinese herbal slices and Chinese patent medicine with reference substance substitution. Methods An HPLC method was used on a Diamonsil C18 column(250 mm×4.6 mm, 5 μm) with the mixture of acetonitrile-0.5 % phosphate solution as mobile phase by gradient elution at the flow rate of 1.0 ml/min. The detection wavelength was 240 nm and the column temperature was 30 ℃. Acetaminophen was selected as the substitute for chlorogenic acid reference substance and the relative correction factor(f) of acetaminophen vs chlorogenic acid was determined by slope method, which was usde to deternine the content of chlorogenic acid in samples. Results The linear ranges of acetaminophen and chlorogenic acid were 5.36-107.19 μg/ml(r=1.0000) and 5.13-102.60 μg/ml(r=0.9999), respectively. The average recoveries of reference substance substitution and external standardization with reference substance were 98.33 % and 98.65 %, respectively, with RSD of 1.34 % and 1.30 %, respectively. The relative correction factor(f) was 0.5119. Different instruments and chromatographic columns, flow rates, column temperature, different determination wavelengths had no significant effects on f value. There were no significant difference between the results of the two quantitative methods. Conclusion HPLC with acetaminophen as the substitute for reference substance of chlorogenic acid is adapted in the quality control of chlorogenic acid in Chinese herbal slices and Chinese patent medicine, which can be applied for the determination of chlorogenic acid.
