两种驱虫方式对阿勒泰羊繁殖母羊体内外寄生虫的驱虫效果及驱虫后对肠道菌群的影响 被引量:3

The effect of two methods for deworming parasites in Aletai ewe and the impact of the treatments on the gut microbiota of the animal

摘要 动物肠道菌群组成结构稳态与宿主健康和疾病关系密切。为了研究伊维菌注射和浇泼两种驱虫方式对绵羊体内体外寄生虫的驱虫效果,以及处理后绵羊肠道菌群的变化,将75只年龄、体重相近的成年健康生产绵羊随机分为3组,每组25只,3个组分别为:不驱虫对照组、浇泼驱虫组、注射驱虫组进行驱虫试验。在驱虫21 d后,评价驱虫效果。每组随机采集15只绵羊直肠粪便样本,采用16S rDNA扩增子技术进行肠道菌群结构分析。结果显示:与对照组相比,浇泼驱虫组和注射驱虫组羊体表寄生虫和体内虫卵在驱虫后均显著降低(P<0.01);2个驱虫组与对照组相比,肠道菌群结构的α和β多样性均无显著差异(P>0.05)。门和属水平物种也未见显著组间差异(P>0.05)。拟杆菌门和厚壁菌门相对丰度较大,为3组羊中的优势菌群。疣微菌科未明确分类的属(Unspecified_Ruminococcaceae)和拟杆菌目未明确分类属(Unspecified_Bacteroidales)为3组羊中的优势菌属。LeFese差异分析表明,经驱虫处理后,绵羊肠道梭菌科(Clostridiaceae)和梭菌属(Clostridium)的丰度显著下降。研究表明,本试验所采用的两种驱虫方式对绵羊均显出良好驱虫效果;驱虫处理对其肠道菌群整体组成结构几乎无改变,而梭菌科、梭菌属相对丰度的显著下降可能对动物健康有益。因此,两种驱虫方式对绵羊正常生理功能未造成不利影响,也间接说明两种驱虫方式的安全性均较好。 The homeostasis of gut microbiota is closely associated with host health and diseases. The impact of two deworming methods, injection and pouring-on with ivermectin preparation against sheep parasites on the gut microbiota was evaluated in ewes. Seventy-five healthy adult breeding ewes with similar body weight were obtained and were randomly divided into three groups, namely, the control, the pouring-on and injection groups, with 25 ewes for each group, to conduct a deworming experiment. Twenty-one days after the experiment, 15 rectal fecal samples from each group were randomly collected to determined gut microbiota using the 16 S rRNA technique. The two methods both showed significant deworming effect against the skin parasites and the eggs of worms in the GI tract. The Alpha and Beta-diversity of gut microbiota didn’t differ between the treated groups. On the phylum level, Firmicutes and Bacteroidetes were the most abundant phyla, which were not significantly different between the three groups. On the genus level,unspecified_Ruminococcaceae and Unspecified_Bacteroidales were two most abundant genuses,which were not significant difference among the three groups. The two deworming methods used in this experiment showed significant effect on Aletai ewes. They had little impact on the gut microbiota,but the significant decrease in the relative abundance of Clostridiaceae and Clostridium after the treatment might be beneficial to the health of the animals. Therefore,the two anthelmintic methods did not adversely affect the normal physiological function of the ewes,which indirectly supported the safety of the two anthelmintic methods tested in this experiment.
