摘要
目的建立安神补脑胶囊HPLC指纹图谱,采用HPLC法测定2,3,5,4'-四羟基二苯乙烯-2-O-β-D葡萄糖苷、淫羊藿苷、甘草酸的质量分数。方法采用ACE Excel C18色谱柱(5μm,4.6 mm×250 mm);乙腈-0.05 mol/L磷酸溶液为流动相,梯度洗脱;流速为1.0 mL/min;柱温为35℃;检测波长为320、270、250 nm。结果50批样品中有13个共有峰,相似度均大于0.96;2,3,5,4'-四羟基二苯乙烯-2-O-β-D葡萄糖苷、淫羊藿苷、甘草酸分别在0.19~190.52μg/mL(r=0.9995),0.20~197.41μg/mL(r=0.9999),0.19~192.47μg/mL(r=0.9999)范围内线性关系良好,平均加样回收率(n=6)分别为100.04%(RSD=1.43%)、97.33%(RSD=1.11%)、97.00%(RSD=1.23%),50批次安神补脑胶囊样品中2,3,5,4'-四羟基二苯乙烯-2-O-β-D葡萄糖苷、淫羊藿苷、甘草酸的质量分数范围分别为2.4~4.8、0.24~1.96、0.04~0.49 mg/粒。结论本文建立的方法简便准确,专属性强,重复性好,可用于安神补脑胶囊的质量控制。
Objective To establish the HPLC fingerprint of Anshen Bunao Capsule and to determine the contents of 2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucoside,Icariin,Glycyrrhizic Acid.Methods The separation of characteristic spectrum was performed on ACE Excel C18 column(4.6 mm×250 mm,5μm)with the mobile phase comprising of acetonitrile-0.05 mol/L phosphoric acid in a gradient elution manner.The flow rate was 1mL/min,the detection wavelengths were set at 320 nm,270 nm,250 nm and the column temperature was 35℃.Results There were thirteen common peaks in the fingerprints of fifty batches of samples with the similarities of more than 0.96.The standard curves were in good linearity in the ranges of 2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucoside 0.19~190.52μg/mL(r=0.9995),Icariin 0.20~197.41μg/mL(r=0.9999),Glycyrrhizic Acid 0.19~192.47μg/mL(r=0.9999),whose average recoveries(n=6)were 100.04%(RSD=1.43%)、97.73%(RSD=1.11%)、97.00%(RSD=1.23%).The contents of 2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucoside,Icariin,Glycyrrhizic Acid in 50 batches of Anshen Bunao Capsules were 2.4~4.8,0.24~1.96,0.04~0.49 mg/grain.Conclusion The method is simple,accurate,specific and reproducible,which can be used for the quality control of Anshen Bunao Capsules.
