摘要
目的观察绿原酸对香烟烟雾提取物(CSE)损伤的人支气管上皮细胞(16HBE细胞)生长增殖和相关细胞因子的影响。方法采用MTT法检测绿原酸的细胞毒性,16HBE细胞用绿原酸孵育后给予CSE刺激,用MTT法测定细胞的生长情况,用ELISA检测细胞上清液中表皮生长因子(EGF)、转化生长因子-β1(TGF-β1)、白细胞介素-10(IL-10)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)的含量,用Western blot法检测细胞外信号调节激酶ERK1/2的表达。结果绿原酸能提高16HBE细胞的相对生长率,提高细胞上清液中EGF、TGF-β1、IL-10的含量,降低TNF-α、IL-6含量,并上调ERK1/2蛋白的表达。结论绿原酸能促进16HBE细胞损伤的修复,该作用可能与介导ERK1/2蛋白表达、促进16HBE细胞的生长增殖、调节生长因子水平、抑制炎症反应有关。
Objective To study the effect of chlorogenic acid on the proliferation and cytokine production of human bronchial epithelial 16HBE cells damaged by cigarette smoke extract(CSE).Methods The cytotoxicity of chlorogenic acid was measured by MTT assay.16HBE cells were treated with or without chlorogenic acid when damaged by CSE.The growth of cells was measured by MTT assay.ELISA was used to detect the levels of EGF,TGF-β1,TNF-α,IL-6 and IL-10.Western blot was used to detect the expression of ERK1/2.Results Chlorogenic acid increased relative growth rate of 16HBE cells,improved the levels of EGF,TGF-β1 and IL-10 and reduced the levels of TNF-αand IL-6 in the supernatant of 16HBE cells.The expression of ERK1/2 was increased in 16HBE cells when treated with chlorogenic acid.Conclusion Chlorogenic acid can promote the repair of 16HBE cells,which may be related to regulation of ERK1/2 expression and cell proliferation,modulation of growth factor and inhibition of inflammatory reaction.
