正交试验法优选五指精蓝颗粒的提取工艺 被引量:2

摘要 [目的]优选五指精蓝颗粒的提取工艺。[方法]通过正交试验,以补骨脂素含量作为水提工艺的观察指标,以人参皂苷Rb1为醇提工艺的观察指标,分别筛选最佳水提工艺和醇提工艺。[结果]以补骨脂素溶液浓度作为横坐标,以峰面积作为纵坐标进行线性回归,得到回归方程为:Y=772 296 357X+81 492,r^2=0.999 7(n=6),表明补骨脂素溶液浓度在0.002~0.020 mg/ml范围内与峰面积线性关系良好,平均加样回收率为99.80%,RSD为0.20%(n=6)。最优水提工艺为:加水量8倍,提取3次,提取时间分别为2 h、1 h、1 h。以人参皂苷Rb1溶液浓度为横坐标,以吸光度为纵坐标,其回归方程为:A=3.565 6C-0.008 9,r^2=0.999 6(n=6),表明人参皂苷Rb1溶液浓度在0.042~0.251 mg/ml范围内与吸光度呈良好的线性关系。最优醇提工艺为:乙醇浓度80%、乙醇量8倍、提取3次,每次2 h。[结论]优选出的醇提工艺和水提工艺简单、合理、稳定性好,提取率较高,可用于工业生产。 [Objective]To optimize the extraction process for wuzhijinglan granules.[Methods]The content of osteoporine was regarded as the observation index of water extraction process,and ginsenoside Rb1 as the observation index of the alcohol extraction process with orthogonal test so as to screen the optimal water and alcohol extraction process respectively.[Results]The concentration of psoralen solution was taken as the x-coordinate and the peak area as the y-coordinate for linear regression.So the regression equation was obtained as Y=772296357 X+81492,r2=0.9997(n=6).The results showed that the concentration of psoralen solution in the range of 0.002~0.020 mg/ml had a good linear relationship with the peak area.The average recovery was 99.80%and the RSD was 0.20%(n=6).The optimal water extraction process was as follows:adding 8 times the amount of water,extracting 3 times,and the extraction time is at 2 h,1 h,and 1 h respectively.With the concentration of ginseng saponin Rb1 solution as the horizontal coordinate and the absorbance as the ordinate coordinate,the regression equation was obtained as A=3.5656 C-0.0089,r2=0.9996(n=6),indicating a good linear relationship between concentration of ginsenoside Rb1 solution in the range of 0.042~0.251 mg/ml and the absorbance.The optimal alcohol extraction process was extraction for 3 times with 80%ethanol concentration and 8 times ethanol volume,2 hours each time.[Conclusion]The optimized alcohol extraction process and water extraction process,which can be used in industrial production,are simple,reasonable,stable,and have a high extraction rate.
