加味左金汤灌胃的SD大鼠血清通过激活细胞线粒体途径促进人胃癌细胞凋亡 被引量:1

Serum of SD rats lavaged by modified Zuojin decoction promotes apoptosis and inhibits proliferation of human gastric cancer cells by activating mitochondrial pathways

摘要 目的观察加味左金汤灌胃的大鼠血清对人胃癌细胞增殖、凋亡的影响及机制。方法采用SD大鼠灌胃加味左金汤,并采集其血清。培养人SGC-7901细胞和MKN-45胃癌细胞,采用(0、25、50、100、200、400)mL/L血清处理胃癌细胞。噻唑蓝(MTT)法观察含药血清对人胃癌细胞增殖的影响,免疫荧光细胞化学染色法检测含药血清处理细胞后KI67抗原(ki67)的表达;流式细胞术检测含药血清对胃癌细胞凋亡的影响,Western blot法检测SGC-7901和MKN-45胃癌细胞裂解型胱天蛋白酶3(c-caspase-3)、c-caspase-8、c-caspase-9、B细胞淋巴瘤因子2(Bcl2)和Bcl2相关X蛋白(BAX)的蛋白水平。结果含药血清可显著抑制SGC-7901和MKN-45细胞的增殖、诱导其凋亡,且ki67表达阳性率显著降低;加味左金汤含药血清处理的胃癌SGC-7901和MKN-45细胞c-caspase-3、c-caspase-9和BAX蛋白水平增加、Bcl2蛋白水平降低。结论加味左金汤含药血清抑制人SGC-7901和MKN-45胃癌细胞的增殖、诱导其凋亡,可能与促进细胞线粒体和死亡受体途径诱导细胞凋亡有关。 Objective To observe the effect of serum of SD rats lavaged by modified Zuojin decoction on the apoptosis and proliferation of human gastric cancer cells and its mechanism.Methods SD rats were gavaged with modified Zuojin decoction to prepare their sera.Human SGC-7901 and MKN-45 cells were cultured and treated with the sera(0,25,50,100,200,400)mL/L.MTT assay was used to observe the effect of drug-containing serum on the proliferation of human gastric cancer cells.Immunofluorescence method was used to detect the expression of ki67 after treatment with the drug-containing serum.The effect of drug-containing serum on the apoptosis of gastric cancer cells was detected by flow cytometry.Western blot analysis was used to detect the protein levels of apoptosis-associated cleaved caspase-3,cleaved caspase-8,cleaved caspase-9,BAX and Bcl2 in SGC-7901 and MKN-45 cells.Results The drug-containing serum significantly inhibited the proliferation and induced the apoptosis of SGC-7901 and MKN-45 cells,and the positive rate of ki67 expression was significantly reduced.The levels of cleaved caspase-3,cleaved caspase-9 and BAX proteins in SGC-7901 and MKN-45 cells increased and the levels of Bcl2 protein decreased.Conclusion The drug-containing serum can significantly inhibit the proliferation and induce the apoptosis of human gastric cancer SGC-7901 and MKN-45 cells,and the mechanism may be related to the activation of mitochondrial pathways.
