电针额区对血管性痴呆大鼠血清NSE、S-100β表达及海马回CA1区细胞凋亡的影响 被引量:1

Effect of Electroacupuncture in the Frontal Area on the Expression of Serum NSE and S-100β and the Apoptosis of Hippocampal CA1 Area in Rats with Vascular Dementia

摘要 目的:观察电针额区对血管性痴呆(VD)模型大鼠血清神经元特异性烯醇化酶(neuron-specific enolase,NSE)、S-100蛋白钙结合蛋白β(S100 calcium binding proteinβ,S-100β)表达及海马回CA1区细胞凋亡的影响。方法:选取60只清洁级健康雄性Wistar大鼠,随机分为假手术组、模型组、西药组和电针组,每组15只。采用2-血管阻断法(2-VO)制作动物模型后,电针组先按照于致顺老师头穴分区法丛刺大鼠额区,再用脉冲电疗仪连接前庭和囟会穴,给予疏密波,20 min后出针,1次/d。西药组大鼠给予脑复康水溶液灌胃,1次/d。模型组与假手术组均灌胃给予生理盐水。然后将4组大鼠进行Morris水迷宫实验,观察治疗30 d后各组大鼠逃避潜伏期及穿越平台次数,并采用酶联免疫吸附法检测大鼠血清NSE、S-100β含量的表达。利用TUNEL法,观察光镜下海马回CA1区细胞凋亡状态。结果:与模型组比较,电针组与西药组逃避潜伏期都明显缩短(P<0.05),穿越平台次数都明显增高(P<0.05),血清NSE及S-100β含量都明显减少(P<0.05),细胞凋亡率都明显降低(P<0.01);电针组逃避潜伏期及穿越平台次数、血清NSE及S-100β含量、细胞凋亡率与西药组比较,差异均无统计学意义(P>0.05)。结论:电针额区能降低VD模型大鼠血清NSE、S-100β含量,减少海马回CA1区细胞凋亡数量,从而提高其学习记忆能力。 Objective: To observe the effect of electroacupuncture in the frontal area on the expression of NSE and S-100β and the apoptosis of hippocampal CA1 area in rats with vascular dementia. Methods: A total of 60 SPF healthy male Wistar rats were randomly divided into sham operation group, model group, western medicine group and electroacupuncture group, with 15 rats in each group. After making the animal model with2-Vascular Occlusion Method(2-VO), the electroacupuncture group first punctured the frontal area of the rats according to the method of YU Zhi-shun’s scalp-point division, and then connected the vestibule and Guihui point with pulse electrotherapy instrument, and gave density wave, which was given once a day after 20 minutes.Rats in the western medicine group were given Naofukang water solution by gavage, once daily. Both the model group and the sham operation group were given normal saline by gavage. Then, Morris water maze test was carried out to observe the escape latency and crossing times of rats in each group after 30 days of treatment, and the expression of NSE and S-100β in serum was detected by enzyme-linked immunosorbent assay. TUNEL method was used to observe the apoptosis of hippocampal CA1 area under light microscope. Results: Compared with the model group, the escape latency of electroacupuncture group and western medicine group were significantly shortened(P<0.05), the times of crossing the platform were significantly increased(P<0.05), the contents of NSE and S-100β in serum were significantly decreased(P<0.05), and the apoptosis rate was also significantly decreased(P<0.01). There were no significant differences in escape latency, crossing platform times, serum NSE and S-100β content, apoptosis rate between the electroacupuncture group and western medicine group(P>0.05).Conclusion: Electroacupuncture in frontal area can reduce the serum NSE and S-100β levels in VD model rats,reduce the number of apoptosis in the CA1 area of the hippocampus, and improving their learning and memory ability.
