摘要
目的:探究榄绿粗叶木多糖的最佳提取工艺,并检测其抗氧化活性和对肝癌细胞株HepG2的抑制活性。方法:以榄绿粗叶木多糖的提取率作为指标,选取超声温度、料液比、超声时间3个因素,采用正交实验优化榄绿粗叶木多糖的提取工艺。通过测定榄绿粗叶木多糖对DPPH自由基的清除能力来评价其抗氧化活性;用0.25、0.50、1.00、2.00、4.00 mg/mL的榄绿粗叶木多糖给药24、48、72 h,考察其对HepG2细胞的抑制作用。结果:多糖提取最佳工艺是超声时间40 min,料液比1∶50,超声温度70℃;在此条件下,榄绿粗叶木多糖的提取率达3.08%,是较为理想的榄绿粗叶木多糖提取工艺;随着多糖浓度的升高,其抗氧化活性升高,清除DPPH自由基的半数清除率IC50为0.092mg/mL;榄绿粗叶木多糖对HepG2肝癌细胞具有抑制增殖作用,在一定范围内,抑制作用随多糖浓度的增加而增加。结论:该方法简单可行,是榄绿粗叶木多糖提取的有效方法。榄绿粗叶木多糖具有一定的抗氧化能力,能抑制HepG2肝癌细胞的生长。
Objective: To explore the best extraction process of the polysaccharides of Lasianthus japonicus,and to detect its antioxidant activity and inhibitory activity on the liver cancer cell line HepG2. Methods: Taking the extraction rate of polysaccharides from Lasianthus japonicus as the index, the extraction process of polysaccharides was optimized by orthogonal experiment with three factors including ultrasonic temperature, solid-liquid ratio and ultrasonic time. The antioxidant activity was evaluated by measuring the scavenging ability of the Lasianthus japonicus polysaccharide on DPPH free radical, and its inhibitory effect on HepG2 cells was investigated by administration of 0.25, 0.50, 1.00, 2.00, 4.00 mg/mL for 24, 48 and 72 h. Results: The optimum extraction conditions were as follows: ultrasonic time 40 min, solid-liquid ratio 1∶50, ultrasonic temperature 70 ℃.Under these conditions, the extraction rate of polysaccharides from Lasianthus japonicus was 3.08%, which was an ideal extraction process. With the increase of polysaccharide concentration, its antioxidant activity increased,and the IC50 of scavenging DPPH free radical was 0.092 mg/mL. The polysaccharides of Lasianthus japonicus can inhibit the proliferation of Hep G2 liver cancer cells. Within a certain range, the inhibitory effect increases with the increase of polysaccharide concentration. Conclusion: The method is simple and feasible, and it is an effective method for extracting polysaccharides of Lasianthus japonicus. The polysaccharides of Lasianthus japonicus has certain antioxidant capacity and can inhibit the growth of Hep G2 liver cancer cells.
