摘要
以‘冷白玉’30 d胚龄的幼胚为材料,对影响胚发育的外源激素、放置方式、胚乳看护时间、光照条件、低温处理的效果进行了研究,以期降低畸形苗发生率,提高幼胚培养效率。结果表明,在IBA0.8 mg/L+ZT0.5 mg/L+GA 37.0 mg/L+NAA0.2 mg/L激素条件下,可获得较高的成胚率56.82%;合点端插入培养基的竖放方式可显著促进幼胚生长发育,幼胚萌发率可达77.39%;胚乳看护适宜时间45 d较为适宜;暗培养利于幼胚正常发育;接种前幼果进行低温处理对减缓畸形苗发生的影响较小。
Young embryos(30-day-old)of Chinese jujube(Ziziphus jujuba‘Lengbaiyu’)were selected as materials.The factors influencing the rescue of the young embryos,including hormones,placement,endosperm care time,light conditions and low temperature were studied in order to reduce the incidence of abnormal seeding and increase the young embryo culture efficiency.The culture condition was optimized as follows:,a higher embryo formation rate of 56.82%could be obtained using the hormones of IBA 0.8 mg/L+ZT 0.5 mg/L+GA 37.0 mg/L+NAA0.2 mg/L;the germination rate of immature embryos could reach 77.39%by putting the chalazal end of embryo into culture medium(radical end up).The appropriate time of endosperm care was 45 days and immature embryos developed normally under the dark condition.0~4℃low temperature before inoculation had little effect on slowing down the occurrence of deformed seedlings.
