中国豫北汉族人群心脑血管疾病患者血管生成素样蛋白4(rs4076317、rs1044250)基因多态性与脂类代谢的关系 被引量:2

Relationship between ANGPTL4 rs4076317,rs1044250 Gene Polymorphism and Lipoprotein Metabolism in Han Chinese from North Henan with Cardiovascular or Cerebrovascular Diseases

摘要 目的探讨分析中国豫北地区汉族心脑血管疾病患者和健康人群血管生成素样蛋白4(ANGPTL4)rs4076317、rs1044250等位基因多态性分布频率、脂类代谢特点,以及rs4076317、rs1044250等位基因与脂类代谢的关系。方法收集2018年12月至2019年12月安阳地区医院住院患者和健康体检人群资料:冠心病组患者(CAD组)143例,缺血性卒中组患者(IS组)130例,健康体检人群(对照组)115例。检测每位研究对象血脂水平;提取每位研究对象血液总DNA,设计rs4076317、rs1044250扩增片段引物,PCR扩增,扩增产物测序;统计分析ANGPTL4 rs4076317、rs1044250等位基因在不同组分布频率有无差异和rs4076317危险度OR值;比较不同组rs4076317、rs1044250不同基因型所对应的血脂水平有无差异,以及不同组血脂水平有无差异。结果(1)CAD组和IS组rs4076317等位基因CC、GC型分布频率与对照组比较,差异有统计学意义(P<0.05);CAD组CC、GC型分布频率和IS组比较,差异无统计学意义(P>0.05)。CAD组、IS组和对照组rs1044250等位基因CC、CT型同类基因型分布频率比较,差异无统计学意义(P>0.05)。CAD组和IS组OR值分别与对照组比较,rs4076317等位基因型CC、GC型危险度OR值分别为0.813、1.709、0.787、1.806,差异有统计学意义(P<0.05)。(2)CAD组、IS组rs4076317基因CC、GC型,rs1044250基因CC、CT型,同类基因型所对应的CHOL、TG、HDL-C、LDL-C水平分别与对照组比较,差异有统计学意义(P<0.05)。CAD组rs4076317 CC、GC型,rs1044250基因CC、CT型,同类基因型对应的CHOL、TG、HDL-C、LDL-C水平分别与IS组比较,差异无统计学意义(P>0.05)。CAD组、IS组和对照组rs4076317基因CC基因型所对应的CHOL、HDL-C、LDL-C水平与GC型分别比较,差异无统计学意义(P>0.05),其TG水平比较,CC型均低于GC型,差异有统计学意义(P<0.05);CAD组、IS组和对照组rs1044250基因CC基因型所对应的CHOL、TG、HDL-C、LDL-C水平与CT型分别比较,差异无统计学意义(P>0.05)。结论中国豫北汉族人群冠心病患者、缺血性卒中患者ANGPTL4 rs4076317等位基因多态性与正常健康人群组分布频率比较有差异,其野生CC型对避免疾病发生具有保护性,杂合突变GC型对疾病发生具有危险性,rs1044250等位基因多态性与正常健康人群组分布频率比较无差异。冠心病患者、缺血性卒中患者ANGPTL4 rs4076317基因多态性与TG代谢指标有关。ANGPTL4 rs4076317等位基因和脂代谢特点,可以作为临床心脑血管疾病风险评估指标,亦对高脂血症病因分析具有一定的帮助。 Objective To investigate the distribution frequency,lipid metabolism characteristics of ANGPTL4 rs4076317,rs1044250 allelic polymorphism in Han population from the Northern region of Henan with cardiovascular and cerebrovascular diseases,and analyze the relationship between rs4076317,rs1044250 alleles and lipid metabolism.Methods Materials of inpatients and health examination population were collected in Anyang District Hospital from December 2018 to December 2019:143 patients with the coronary heart disease(CHD),130 patients with ischemic stroke(IS),and 115 healthy persons for control group were recruited.Serum lipid content of every subject was determined,and total DNA was extracted from the blood of every subject.The primers of ANGPTL4 rs4076317 and rs1044250 were designed and PCR amplification was performed,the PCR production were sequenced.The distribution frequency difference of ANGPTL4 rs4076317 and rs1044250 allele in different groups were statistically analyzed,and the odds ratio(OR)of rs4076317 was statistically analyzed.The contents of serum lipid of different genotypes of ANGPTL4 rs4076317and rs1044250 in different groups and the contents of serum lipid in different groups were compared.Results(1)There were statistical differences in genotype CC,GC frequencies of rs4076317 in CAD group and IS group compared with them in control group(P<0.05).There was no statistical difference in genotype CC,GC frequencies of rs4076317 between CAD group and IS group(P>0.05).There was no statistical difference in genotype CC,GC frequencies of rs1044250 among CHD group,IS group and control group(P>0.05).The CC,GC genotypes of rs4076317 of CAD group and IS group were compared with the control group,and the OR values were 0.813,1.709,0.787,1.806 respectively(P<0.05).(2)There were statistical differences in levels of CHOL,TG,HDL-C,LDL-C of CC,GC of rs4076317,CC,CT of rs1044250 genotype which the same genotype in CAD group and IS group compared with them of control group(P<0.05).There was no statistical difference in levels of CHOL,TG,HDL-C,LDL-C of CC,GC of rs4076317,CC,CT of rs1044250 which the same genotype in CAD group compared with IS group(P>0.05).There was no statistical difference in levels of CHOL,HDL-C,LDL-C of rs4076317 CC genotype compared with them of the GC genotype in all groups(P>0.05),while the TG levels of rs4076317 CC genotype were lower than GC genotype in all groups(P<0.05).There was no statistical difference in levels of CHOL,TG,HDL-C,LDL-C of rs1044250 CC genotype compared with that CT genotype in all groups(P>0.05).Conclusion The distribution frequencies of ANGPTL4 rs4076317 allele polymorphism in CHD and IS patients are different from them in healthy persons in Han Chinese from North Henan.The wild CC genotype is protective to avoid disease occurrence,while heterozygous mutation GC type is dangerous to disease occurrence.There is no statistical difference in the distribution frequencies of ANGPTL4 rs1044250 allele polymorphism in CHD and IS patients compared with them in healthy persons.The polymorphism of ANGPTL4 rs4076317 gene are associated with TG metabolism in CHD and IS patients.ANGPTL4 rs4076317 alleles and lipid metabolism characteristics can be used as risk assessment indicators for clinical cardiovascular and cerebrovascular diseases,and can also be helpful for the etiological analysis of hyperlipidemia.
