摘要
为分析克里曼丁红橘果皮色泽变异的分子机理,本研究以克里曼丁红橘橙黄色芽变植株为材料,利用SRAP分子标记对其进行分析,100对引物中,成功扩增并克隆得到了2条差异片段。其中从黄色克里曼丁样品中扩增出一条长474 bp的差异片段,与甜橙基因组中Cs1g22940.1部分序列完全一致;另外从红色克里曼丁样品中扩增出一条长323 bp的差异片段,与甜橙基因组中Cs4g12370部分序列完全一致。取12个不同果皮色泽的柑橘品种对两对引物的扩增结果进行验证,结果表明,323 bp的差异片段更有可能与果皮色泽相关。本研究为分析柑橘果皮色泽差异提供了科学依据。
In order to analyze the molecular mechanism of color variation of clementine peel, this study took Clementine mandarin orange bud transformation plant as the material and analyzed it with SRAP molecular marker,and successfully amplified in 100 pairs of primers. Two differential fragments were successfully amplified and cloned. Among them, one differential fragment of 474 bp was amplified from the yellow Clementine sample,which was identical to the sequence of Cs1g22940.1 in the orange genome, and the other differential fragment of long 323 bp was amplified from red Clementine sample, which was completely consistent with the sequence of Cs4g12370 in orange genome. The amplification results of the two pairs of primers were verified with 12 citrus varieties with different peel color. The results showed that the 323 bp difference fragment was more likely to be related to peel color. This study provides a working basis for analyzing the difference in color of citrus peel.
