玫瑰红芪当归水煎剂对围绝经期综合征大鼠性激素水平及氧化指标的影响 被引量:7

摘要 目的:观察玫瑰红芪当归水煎剂对围绝经期综合征大鼠内分泌激素含量及SOD活性的影响并探讨其治疗机制。方法:通过手术摘除Wistar雌性大鼠双侧卵巢复制围绝经期综合征模型,血清雌二醇(estradiol,E 2)含量检测以确定模型成功后,将其随机分为假手术组、卵巢切除组、玫瑰红芪当归水煎剂低、中、高剂量组(1.4、2.8、5.6g/kg)和戊酸雌二醇片组,连续干预6周。处死大鼠后测量子宫质量,HE染色观察各组大鼠子宫组织形态学变化,比色分析法测定各组大鼠子宫组织超氧化物歧化酶(superoxide dismutase,SOD)活性和丙二醛(malondialdehyde,MDA)含量,ELISA法测定大鼠血清雌二醇(estradiol,E 2)、卵泡刺激素(follicle-stimulating hormone,FSH)、促黄体生成激素(luteinizing hormone,LH)的含量。结果:与卵巢切除组比较,玫瑰红芪当归水煎剂中、高剂量组和戊酸雌二醇片组可有效降低卵巢切除模型大鼠体质量、MDA含量、FSH含量、LH含量,升高卵巢切除模型大鼠子宫指数和SOD活性,玫瑰红芪当归水煎剂低、中、高剂量和戊酸雌二醇片能有效升高卵巢切除模型大鼠E 2的含量。HE染色结果发现,玫瑰红芪当归水煎剂高剂量组和戊酸雌二醇片组子宫黏膜层出现腺体,单层柱状上皮细胞排列整齐。结论:玫瑰红芪当归水煎剂可通过调节围绝经期综合征大鼠抗氧化功能和内分泌功能延缓卵巢的衰老进程,可能是其治疗女性围绝经期综合征的作用机制之一。 Objective:To investigate the effect of Rosa rugosa,Radix Hedysari and Angelicae Sinensis Radix decoction on the endocrine hormone content and SOD activity in perimenopausal syndrome rats,which was to explore its therapeutic mechanism.Methods:Rat model of perimenopausal syndrome was induced by bilateral ovariectomy.The models were replicated successfully by the E2 level of serum,they were randomly divide into sham operation group,ovariectomy group,Rosa rugosa,Radix Hedysari and Angelicae Sinensis Radix decoction low-dose,medium-dose and high-dose groups.After 6 weeks’intragastrical administration,the uterine weights were measured,the morphology changes of uterus tissue were observed by HE,the levels of SOD and MDA in uterus tissue were checked by colorimetric assay,the concentrations of E2,FSH,and LH in serum were measured by ELISA.Results:Compared with the ovariectomy group,the rat weights and the levels of serum MDA,FSH,LH were all decreased in Rosa rugosa,Radix Hedysari and Angelicae Sinensis Radix decoction medium-dose,high-dose group and estradiol valerate tablet group,the uterine coefficient and the SOD activity in uterine were increased in Rosa rugosa,Radix Hedysari and Angelicae Sinensis Radix decoction medium-dose,high-dose group and estradiol valerate tablet group,the E2 content was also increased in Rosa rugosa,Radix Hedysari and Angelicae Sinensis Radix decoction treatment group and estradiol valerate tablet group.HE staining showed that the glands appeared and monolayer columnar epithelial cells arranged neatly in Rosa rugosa,Radix Hedysari and Angelicae Sinensis Radix high-dose group and estradiol valerate tablet group.Conclusion:The therapy of Rosa rugosa,Radix Hedysari and Angelicae Sinensis Radix decoction can delay the aging process of the ovary by regulating the antioxidant and the endocrine function in ovariecomized rats,which may be one of the mechanisms in the treatment of female perimenopausal syndrome.
