摘要
以果胶为载体,用二甲基十八烷基[3-(三甲氧基硅基)丙基]氯化铵为反应活性抗菌单体制备果胶季铵盐衍生物,利用果胶与有机硅季铵盐的交联反应,给果胶分子表面固载抗菌基团,为扩大果胶的应用范围开辟新的途径。用红外光谱、1H NMR、Zeta电位仪和扫描电子显微镜(SEM)对产物进行了表征,用悬菌定量实验研究了其抗菌性能。结果表明:二甲基十八烷基[3-(三甲氧基硅基)丙基]氯化铵通过化学键可以固载到果胶表面形成果胶季铵盐衍生物;自制橘皮果胶的零电位为pH≈3.4,而所制备的果胶季铵盐衍生物的零电位则为pH≈8.2,在果胶分子中引入季铵基团致其零电位显著提高;所制备的果胶季铵盐衍生物对金黄色葡萄球菌、大肠杆菌与白色念珠菌均有良好的杀菌作用,接触30 min的杀菌率均为99.99%。用此方法能够制备出具有良好抗菌活性的果胶季铵盐衍生物抗菌材料,可以为拓展果胶的使用范围、提高其经济效益提供参考。
The quaternary ammonium salt derivatives of pectin were prepared with pectin as carrier and dimethyl octadecyl[3-(trimethoxysilyl)propyl]ammonium chloride as active antibacterial monomer.The cross-linking reaction between pectin and organosilicon quaternary ammonium salt was conducted to immobilize antibacterial groups on the surface of pectin molecules,which broadened the application of pectin.The pectin derivative was characterized by Fourier transform infrared spectroscopy(FTIR),1H NMR,Zeta sizer nano series,and scanning electron microscope(SEM).The antimicrobial properties of the pectin derivative against selected microorganisms were tested by the quantitative suspension method.The results revealed that(1)N-Octadecyl dimethyl(3-trimethoxysilylpropyl)ammonium chloride can be fixed onto the surface of pectin through chemical bonds.(2)The zero potential of the pectin and the pectin derivative was pH 3.4 and pH 8.2,respectively,and the quaternary ammonium groups significantly increased the zero potential.(3)The pectin derivative significantly inhibited the growth of E.coil,S.aureus and C.albicans with the inhibition rate of 99.99%for 30 min.In conclusion,quaternary ammonium salt derivatives of pectin with good antibacterial activity can be prepared with this method,which provides a reference for expanding the application of pectin and improving its economic benefits.
