忍冬ICE1基因密码子偏好性分析及受体系统选择 被引量:5

Codon bias analysis and receptor system selection of ICE1 gene in Lonicera japonica Thunb.

摘要 为揭示忍冬(Lonicera japonica) ICE1基因密码子使用特性,运用CodonW软件、EMBOSS在线程序分析了其密码子的使用偏好性,与山茶等8种植物的ICE1基因的密码子偏好性进行比较,并进行中性绘图、ENc分析和奇偶偏好偏差分析。结果表明:忍冬ICE1基因密码子偏好以A/T结尾,且密码子使用偏好性较弱;RSCU值分析确定忍冬ICE1基因中有27个高频密码子;系统聚类发现,基于CDS的系统聚类比基于RSCU值的系统聚类更加准确;碱基成分和相关性分析发现ICE1密码子除突变影响外,选择效应也产生了一定的影响;密码子使用频率分析发现,拟南芥和烟草均可作为忍冬ICE1基因的遗传转化受体,酵母菌表达系统较大肠杆菌更适合忍冬ICE1基因的异源表达。忍冬ICE1基因密码子偏好性分析及瞬时表达验证,能够为该基因的异源表达和分子遗传研究提供一定的基础。 In order to reveal the codon usage characteristics of ICE1 gene in Lonicera japonica, the codon usage was analyzed by Codon W software and EMBOSS online program, compared codon bias of ICE1 gene with other 8 plants such as Camellia japonica. Neutral plotting, ENc analysis, and parity bias analysis were performed. The results showed that the codon bias of ICE1 gene was ended with A/T and the codon bias was low;according to the RSCU value, it was determined 27 high frequency codons of the ICE1 gene in Lonicera japonica;systematic clustering found that clustering based on CDS is more accurate than clustering based on RSCU values;The codon base composition and correlation analysis showed that in addition to the mutation effect of the ICE1 codon, the selection effect also had a certain effect;The codon usage frequency analysis showed that both Arabidopsis thaliana and Nicotiana tabacum could be used as genetic transformation receptors for the ICE1 gene of Lonicera japonica. The yeast expression system is more suitable for heterologous expression of the LjICE1 gene than E. coli. The codon bias analysis and instantaneous expression verification of LjICE1 gene can provide a certain basis for the heterologous expression and molecular genetic research of the ICE1 gene in Lonicera japonica.
