化湿汤对溃疡性结肠炎模型大鼠的治疗作用及机制研究

摘要 目的观察化湿汤对溃疡性结肠炎(UC)大鼠的治疗作用,并从抗炎、抗氧化损伤角度探讨其作用机制。方法将50只SD采用三硝基苯磺酸(TNBS)灌肠制作UC大鼠模型,造模成功后将大鼠随机分为模型组、阳性药物组、化湿汤低剂量组、化湿汤中剂量组、化湿汤高剂量组,每组10只,另设空白组大鼠10只。各组大鼠均按10 m L/kg灌胃,空白组及模型组予蒸馏水灌胃,阳性药物组予0. 036 g/m L美沙拉秦缓释颗粒药液灌胃,化湿汤低、中、高剂量组分别予化湿汤0. 5、1. 0、2. 0 g/m L药液灌胃,每日1次,连续灌胃10 d。观察各组大鼠造模后第1、3、6、9、12、15 d的体质量、毛色、粪便性状等一般情况。比较各组大鼠末次灌胃后血清白细胞介素1β(IL-1β)、IL-10、一氧化氮(NO)水平及结肠组织中丙二醛(MDA)、超氧化物歧化酶(SOD)、髓过氧化物酶(MPO)水平变化情况。结果与空白组比较,模型组大鼠的体质量在造模后第3、6、9、12及15 d均显著降低(P <0. 05);与模型组比较,阳性药物组及化湿汤中、高剂量组造模后第15 d体质量均明显升高(P <0. 05);与化湿汤低剂量组比较,化湿汤高剂量组大鼠造模后第15 d体质量明显升高(P <0. 05)。与空白组比较,模型组大鼠血清IL-1β和NO水平明显升高(P <0. 05),IL-10水平明显下降(P <0. 05);与模型组比较,阳性药物组及化湿汤高剂量组大鼠血清IL-1β和NO水平明显降低(P <0. 05),IL-10水平明显升高(P <0. 05),化湿汤中剂量组大鼠血清IL-1β和NO水平降低(P <0. 05);与化湿汤低剂量组比较,化湿汤高剂量组血清IL-10水平升高(P <0. 05),NO水平降低(P <0. 05),化湿汤中剂量组仅血清NO降低(P <0. 05)。与空白组比较,模型组大鼠结肠组织中MDA和MPO水平均明显升高(P <0. 05),SOD水平明显降低(P <0. 05);与模型组比较,化湿汤中、高剂量组大鼠结肠组织中MDA和MPO水平均明显降低(P <0. 05),SOD水平均明显升高(P <0. 05),阳性药物组MPO水平降低(P <0. 05);与化湿汤低剂量组比较,化湿汤高剂量组结肠组织中MDA水平降低(P <0. 05),SOD水平升高(P <0. 05),而化湿汤中剂量组仅MDA水平降低(P> 0. 05)。结论化湿汤治疗UC模型大鼠疗效确切,可明显改善大鼠临床症状,改善病变结肠组织形态,其作用机制可能与抑制炎性反应,减轻氧化损伤有关,且高剂量效果更好。 Objective To observe the therapeutic effects of Huashi decoction(HSD)on ulcerative colitis(UC)model in rats and to explore its mechanism from the perspective of anti-inflammation and anti-oxidative damage.Methods Fifty SD rats with UC model were prepared successfully for the enema with 2,4,6-trinitrobenzenesulfonic acid(TNBS)and were randomly divided into the model group(n=10)for the enema with distilled water(10 mL/kg,qd),the positive drug group(n=10)with mesalazine SR granules liquid(0.036 g/mL and 10 mL/kg,qd),the low-dose HSD group(n=10)with HSD(0.5 g/mL and 10 mL/kg,qd),the medium-dose HSD group(n=10)with HSD(1.0 g/mL and 10 mL/kg,qd),and the high-dose HSD group(n=10)with HSD(2.0 g/mL and 10 mL/kg,qd).Another 10 rats were selected into the control group with distilled water(10 mL/kg,qd).The enema period was 10 days.The rat body mass,hair color,and fecal character were observed at Day 1,3,6,9,12 and 15 after the modeling.After the last enema administration,the levels of interleukin-1β(IL-1β),interleukin-10(IL-10),and nitric oxid(NO)in serum and the levels of malonaldehyde(MDA),superoxide dismutase(SOD),and myeloperoxidase(MPO)in colon tissues were compared.Results In comparison to the results of the control group,the body mass index for rats in the model group was significantly lower(P<0.05)at Day 3,6,9,12 and 15 after the modeling;the improved levels of IL-1βand NO were obviously higher(P<0.05)while the improved level of IL-10 was obviously lower(P<0.05);the improved levels of MDA and MPO were obviously higher(P<0.05)while the improved level of SOD was obviously lower(P<0.05).In comparison to the results of the model group,the body mass indexes for rats in the positive drug group,the medium-dose HSD group,and the high-dose HSD group were significantly higher(P<0.05)at Day 15 after the modeling;the improved levels of IL-1βand NO in the positive drug group and the high-dose HSD group were obviously lower(P<0.05)while the improved level of IL-10 was obviously higher(P<0.05);the improved levels of IL-1βand NO in the medium-dose HSD group was significantly lower(P<0.05);the improved levels of MDA and MPO in the medium-dose HSD group and the high-dose HSD group were obviously lower(P<0.05)while the improved level of SOD was obviously higher(P<0.05);the improved level of MPO in the positive drug group was obviously higher(P<0.05).In comparison to the results of the low-dose HSD group,the body mass index for rats in the high-dose HSD group were significantly higher(P<0.05)at Day 15 after the modeling;the improved levels of IL-1βand NO in the high-dose HSD group were higher(P<0.05);the improved levels of NO in the medium-dose HSD group were higher(P<0.05);the improved levels of MDA and SOD in the high-dose HSD group were obviously higher(P<0.05)while the improved level of MDA in the medium-dose HSD group was obviously higher(P>0.05).Conclusion HSD on UC model in rats has a definite therapeutic effect,which can improve the clinical symptoms and the histomorphological changes of diseased colon in rats.Its mechanism may be related to inhibiting the inflammatory reaction and relieving the oxidative damage.And the high-dose HSD treatment has a better effect.
