摘要
目的探讨尿路致病性大肠埃希菌(UPEC)的量与尿路感染模型大鼠尿道上皮组织细胞毒性坏死因子(CNF)1蛋白及CD36蛋白表达水平的关系。方法将50只6~8周龄雌性SD大鼠作为研究对象,随机选取10只作为对照组,其余40只用于感染不同梯次的UPCE建立尿路感染模型,分别为10^1CFU组、10^2CFU组、10^3CFU组和10^4CFU组,每组10只。感染5 d后,定量大鼠尿道上皮组织与尿液菌量,qRT-PCR检测CNF1 mRNA和CD36 mRNA水平,Western印迹检测CNF1蛋白和CD36蛋白表达水平。结果各组尿道上皮组织和尿液菌量均有显著差异(P<0.05),在10^1CFU组、10^2CFU组、10^3CFU组和10^4CFU组中,尿道上皮组织和尿液菌量依次升高,两两比较差异均有统计学意义(P<0.05)。对照组、10^1CFU组、10^2CFU组、10^3CFU组和10^4CFU组中,CNF1和CNF1 mRNA表达水平依次升高,CD36和CD36 mRNA表达水平依次降低,两两比较差异均有统计学意义(P<0.05)。尿道上皮组织和尿液菌量与CNF1表达呈正相关(P<0.05),与CD36表达呈负相关(P<0.05)。结论感染大鼠尿路菌量越高,其尿道上皮组织菌量、尿液菌量和CNF1表达越高,CD36表达越低,尿道感染可能与高菌量影响CNF1和CD36表达相关。
Objective To investigate the relationship between the amount of urinary tract pathogenic escherichia coli(UPEC)and the levels of cytotoxic necrosis factor(CNF)1 protein and macrophage CD36 protein in rats epithelial tissues of urethral infection model.Methods 50 female SD rats aged 6~8 weeks were selected as the study subjects,10 rats were randomly selected as the control group,and the remaining 40 rats were used for the UPCE with different ladders to establish the model of urethral infection,which were 10^1 CFU group,10^2 CFU group,10^3 group and 10^4 group,with 10 rats each group.After 5 days of infection,CNF1 mRNA and CD36 mRNA levels of the rats urethral tissue and urine bacteria volume were quantified,detected by qRT-PCR,Western blot was used to detect CNF1 protein and CD36 protein expression levels of urethral tissue.Results There were significant differences in the volume of urethral tissue and urine bacteria among the five groups(P<0.05).In the 10^1 CFU,10^2 CFU,10^3 CFU and 10^4 CFU groups,the volume of urethral tissue and urine bacteria increased in turn,and the differences between the two groups were statistically significant(P<0.05).CNF1,CNF1 mRNA,CD36 and CD36 mRNA expressions of urethral tissue were significantly different in five groups of rats(P<0.05),in the control,10^1 CFU,10^2 CFU,10^3 CFU and 10^4 CFU groups,CNF1 and CNF1 mRNA expressions were increased in turn,CD36 and CD36 mRNA expressions were decreased in turn,and the differences between the two comparisons were statistically significant(P<0.05).The volume of urethral tissue and urine bacteria were positively correlated with CNF1 expression(P<0.05)and negatively correlated with CD36 expression(P<0.05).Conclusions The higher volume of urethral bacteria in infected rats,the higher volume of urethral tissue bacteria,urine bacteria and CNF1 expression,and the lower CD36 expression,urethral infection might be related to the influence of high volume of bacteria on CNF1 and CD36 expression.
