摘要
目的:探讨湿润暴露疗法/湿润烧伤膏(MEBT/MEBO)对慢性创面组织信号转导和转录激活子6(STAT6)、细胞因子信号转导抑制蛋白1(SOCS1)表达的影响。方法:取SPF级Wistar雄性大鼠54只,随机分为美宝组、贝复新组和模型组,每组18只。构建慢性难愈合创面大鼠模型,分别使用美宝、贝复新和生理盐水纱布覆盖创面。记录3组创面愈合时间及不同时点的创面愈合率,建模后第3、7、14天观察创面组织病理变化,实时定量PCR及免疫印迹法检测创面组织STAT6、SOCS1 mRNA及蛋白表达。结果:美宝组和贝复新组创面愈合时间均短于模型组(均P<0.05)。建模后第7天、第14天,美宝组和贝复新组创面愈合率均高于模型组(均P<0.05)。建模后第7天、第14天,Masson染色显示美宝组和贝复新组创面愈合情况优于模型组。免疫印迹及实时定量PCR显示,建模后第3天、第7天,美宝组和贝复新组STAT6 mRNA及蛋白表达均低于模型组,SOCS1 mRNA及蛋白表达均高于模型组(均P<0.05);建模后第14天,美宝组和贝复新组STAT6蛋白表达低于模型组(均P<0.05),STAT6 mRNA、SOCS1 mRNA及蛋白表达与模型组比较差异无统计学意义(均P>0.05)。结论:MEBT/MEBO可能通过调控STAT6、SOCS1表达而促进慢性创面愈合。
Objective:To investigate the effects of moist exposed burn therapy/moist exposed burn ointment(MEBT/MEBO)on the expression levels of signal transducer and activator of transcription 6(STAT6)and suppressor of cytokine signaling 1(SOCS1)in chronic wound tissues.Methods:Fifty-four male SPF Wistar rats were included in the study and randomly divided into the MEBO group,rb-bFGF group and model group(n=18 each).A rat model of chronic refractory wounds was established,and the wounds were covered with MEBO,rb-bFGF and saline-gauze dressings.The time to wound healing and wound healing rate at different time points in the 3 groups were recorded.The pathological changes of wound tissues were examined at 3,7 and 14 d after modeling.Real-time quantitative PCR and western blotting were used to determine the mRNA and protein expression levels of STAT6 and SOCS1 in wound tissues.Results:The time to wound healing in the MEBO group and rb-bFGF group was shorter than that in the model group(both P<0.05).At 7 and 14d after modeling,the wound healing rate in the MEBO group and rb-bFGF group was higher than that in the model group(all P<0.05).At 7 and 14d after modeling,Masson staining showed that the wound healing in the MEBO group and rb-bFGF group was better than that in the model group.Western blotting and real-time quantitative PCR showed that the mRNA and protein expression levels of STAT6 in MEBO group and rb-bFGF group were lower than those in the model group at 7 and 14d after modeling,whereas the mRNA and protein expression level of SOCS1 was higher than that in the model group(all P<0.05).At 14d after modeling,the STAT6 protein expression level in the MEBO group and rb-bFGF group was lower than that in the model group(all P<0.05),and there were no statistically significant differences in the STAT6 mRNA,SOCS1 mRNA and protein expression levels compared with those in the model group(all P>0.05).Conclusion:MEBT/MEBO may promote chronic wound healing by regulating the expression levels of STAT6 and SOCS1.
