摘要
目的研究不同低温保存液对大鼠离体腹主动脉血管功能及结构的影响。方法选取成年SD大鼠腹主动脉作为研究对象,分别用威斯康星大学保存液(University of Wisconsin solution,UW)、组氨酸色氨酸酮戊二酸酯(histidine-tryptophanketoglutarate solution,HTK),0.9%氯化钠注射液(normal saline,NS)及乳酸钠林格氏液(Ringer's solution,RS)作为保存剂于4℃冰箱中冷藏保存,用离体血管张力测定实验检测血管功能,MTT比色法检测血管组织剩余细胞活性,病理切片评估血管骨架情况,病原学培养评估其安全性。结果1)病原学培养结果:保存1个月内,仅RS组细菌培养阳性,保存3个月后,各组均培养出细菌。2)组织病理结果:保存1个月内,各组血管管壁各层结构均逐渐退变,各保存组间变化无明显差异;保存3个月时,NS组和RS组血管退变最为明显,胶原纤维及弹性纤维可见溶解断裂现象。3)MTT实验结果:保存3 d内,各保存组效果相当(P>0.05);保存14~30 d,UW组保存效果优于其他三组(P<0.05);保存30 d后,各组保存效果均明显下降,差异无统计学意义(P>0.05)。4)张力实验结果:保存3 d内,NS组保存效果最佳(P<0.05);保存7~14 d,UW组与HTK组保存效果相当(P>0.05),均优于另外两组(P<0.05);保存14 d后,UW组保存效果最佳(P<0.05)。结论1个月内各组保存液均能较好地保存离体血管骨架结构,但UW液能更好地保存离体血管细胞活性和收缩功能。
Objective To explore the effects of different cryopreservation solutions on the function and structure of isolated blood vessels.Methods The abdominal aorta of adult SD rats was selected as the research object,the University of Wisconsin solution(UW group),histidine-tryptophan-ketoglutarate solution(HTK group),sterile normal saline(NS group)and Ringer's solution(RS group)were used as preservatives.The function of blood vessel was detected by tension test in vitro,the activity of residual cells in vascular tissue was detected by MTT colorimetry,the vascular skeleton was evaluated by pathological section,and the pathogen culture was evaluated for the safety.Results Pathogen culture results showed that within 1 month,only the RS group had positive result in bacteriological culture,but bacteria could be cultured in each group after 3 months of storage.Histopathological results showed that within 1 month,the structure of each layer of vascular wall in each group gradually degenerated,and there was no significant difference among the preservation groups.After three months of storage,the vascular degeneration was most obvious in the NS and the RS groups,and collagen fibers and elastic fibers were dissolved and broken.MTT test results suggested that within 3 days,the effect of each preservation group was similar(P>0.05);the protective effect of UW group was better than the other three groups(P<0.05);after 30 days of storage,the protective effect of each group decreased significantly,while the difference was not statistically significant(P>0.05).Tension test results showed that within 3 days,the NS group had the best performance(P<0.05);after 7-14 days,the UW group and the HTK group had the same effect(P>0.05),which were better than the other two groups(P<0.05);after 14 days of storage,UW group had the best effect(P<0.05).Conclusion Within one month,UW solution can preserve the vascular cytoskeleton in vitro,but UW solution can better preserve the viability and contractile function of isolated vascular cells.
