摘要
目的:探究阳和汤对实验性自身免疫性甲状腺炎(EAT)大鼠的治疗效果。方法:24只雌性SD大鼠随机分为正常组、模型组、阳和汤治疗组、阳和汤预防组。除正常组外,其余各组运用猪甲状腺球蛋白和弗氏佐剂混合免疫注射法,联合高碘喂养法,建立EAT大鼠模型。阳和汤治疗组造模完成后给予阳和汤灌胃,阳和汤预防组造模同时阳和汤灌胃处理。通过酶联免疫法检测大鼠血清中甲状腺过氧化酶抗体(TPO-Ab)、甲状腺球蛋白抗体(TgAb)和IL-35水平,取完整的甲状腺组织进行HE染色,观察大鼠甲状腺病理学形态变化,分析评价阳和汤治疗效果。结果:(1)与正常组比较,模型组大鼠血清中TPO-Ab和TgAb表达明显升高,IL-35表达明显下降,甲状腺滤泡萎缩,可见大量淋巴细胞炎症浸润。(2)与模型组比较,阳和汤治疗组大鼠血清中TPO-Ab和TgAb表达明显降低,IL-35表达明显升高,甲状腺滤泡形态较规则,淋巴细胞炎症浸润有明显改善。而阳和汤预防组大鼠血清中TPO-Ab、TgAb和IL-35表达及甲状腺组织形态无明显改善。(3)EAT大鼠模型中IL-35表达与TPO-Ab表达水平呈密切负相关。(4)阳和汤可抑制EAT大鼠PBMC中STAT3通路激活。结论:阳和汤可以改善自身免疫性甲状腺炎大鼠的甲状腺功能,有效降低EAT大鼠血清中甲状腺抗体水平,缓解甲状腺炎症状,但是不能预防自身免疫性甲状腺炎的发生。
Objective: To explore the therapeutic effect of Yanghe Decoction on experimental autoimmune thyroiditis(EAT) rats. Methods: A total of 24 female SD rats were randomly divided into normal group, model group, Yanghe Decoction treatment group, and Yanghe Decoction prevention group. Except for the normal group,the other groups used porcine thyroglobulin and Freund’s adjuvant mixed immunization injection method, combined with high iodine feeding method to establish an EAT animal model. Yanghe Decoction treatment group was given Yanghe Decoction by gavage after the completion of the model, while Yanghe Decoction prevention group was given Yanghe Decoction by gavage at the model was being built. The levels of thyroid peroxidase antibody(TPO-Ab), thyroglobulin antibody(TgAb) and IL-35 in rat serum were detected by enzyme-linked immunosorbent assay(ELISA). The intact thyroid tissues was taken for HE staining to observe the pathological changes of thyroid gland in rats, and to analyze and evaluate the therapeutic effect of Yanghe Decoction. Results:(1) Compared with the normal group, the expression of TPO-Ab and TgAb in serum of the model group was significantly increased,the expression of IL-35 was significantly decreased, the thyroid follicle was atrophy, and a large number of lymphocytes inflammatory infiltration were observed.(2) Compared with the model group, the expression of TPO-Ab and TgAb in serum of rats in the Yanghe Decoction treatment group was significantly reduced, the expression of IL-35 was significantly increased, the morphology of thyroid follicle was more regular, and the inflammatory infiltration of lymphocytes was significantly improved. However, the expression of TPO-Ab, TgAb and IL-35 in serum and the morphology of thyroid tissue were not significantly improved in Yanghe Decoction prevention group.(3) There was a close negative correlation between IL-35 expression and TPO-Ab expression in the EAT rats.(4) Yanghe Decoction can inhibit the activation of STAT3 pathway in PBMC of EAT rats. Conclusion:Yanghe Decoction can improve the thyroid function of rats with autoimmune thyroiditis, which can effectively reduce the level of thyroid antibody in serum of EAT rats, and relieve the symptoms of thyroiditis, but it cannot prevent the occurrence of autoimmune thyroiditis.
