摘要 目的探究百合枣仁袋泡茶的制备工艺,建立简单、快速、灵敏同时测定百合枣仁袋泡茶中酸枣仁皂苷A、B和斯皮诺素的HPLC-MS方法。方法采用MRM多反应监测负离子模式,Welch Ultimate XB-C18(2.1 mm×50 mm,5μm)色谱柱,0.1%甲酸水-乙腈梯度洗脱,流速为0.60 mL/min,柱温30℃。结果酸枣仁皂苷A、B和斯皮诺素在5~1000 ng/mL的浓度范围内,线性关系良好;平均回收率为95.83%~100.02%,RSD值在3.0%~7.8%之间;10批百合枣仁袋泡茶的酸枣仁皂苷A、酸枣仁皂苷B、斯皮诺素含量分别为(102.92±2.60)μg/g、(49.81±1.12)μg/g和(205.28±5.95)μg/g,其RSD值分别为3.86%、2.25%和2.90%。结论该法专属性强、快速灵敏、操作简单快捷,可用于百合枣仁袋泡茶中酸枣仁皂苷A、B和斯皮诺素含量的测定。 Objective To study the preparation technology of Lily Semen ziziphi spinosae tea bag and establish a method for simultaneous determination of 3 components including Jujuboside A,Jujuboside B and Spinosin in the tea bag by HPLC-MS.Methods The column was Welch Ultimate XB-C18(2.1 mm×50 mm,5μm),mobile phase consisted of 0.1%formic acid water-acetonitrile at a flow rate of 0.6 mL/min with a 15 minutes gradient elution program.The temperature of column was 30℃.The HPLC-MS/MS system was operated by using an electrospray ionization probe in the negative ion mode.Scan mode was in multiple reaction ion monitoring(MRM)mode.Results The linear ranges of jujuboside A,jujuboside B and spinosin were 5-1000 ng/mL.The average recovery was 95.83%-100.02%and RSD was 3.0%-7.8%.The average content of jujuboside A,jujuboside B and spinosin in ten batches of Lily Semen ziziphi spinosae tea bag was(102.92±2.60)g/g,(49.81±1.12)g/g and(205.28±5.95)g/g,respectively.Their RSD were 3.86%,2.25%and 2.90%,respectively.Conclusion A sensitive,accuracy and suitable HPLC-MS method has been developed,and the method can be applied for the detemination of jujuboside A,jujuboside B and spinosin in Lily Semen ziziphi spinosae tea bag.
