摘要
目的:建立沙棘籽油微乳维生素E的含量测定方法,优化沙棘籽油微乳处方组成及其配比。方法:采用HPLC测定沙棘籽油在不同油相、乳化剂、助乳化剂中的溶解度;观察滴入水相后体系的外观形态变为澄清时的加水量,绘制伪三元相图,获得最佳处方组成;以乳化剂、油相和助乳化剂的用量百分比为自变量,沙棘籽油微乳中维生素E含量和沙棘籽油微乳的粒径为响应值,采用Box-Behnken响应面法优化沙棘籽油微乳最佳处方配比。结果:沙棘籽油微乳最佳处方组成及最佳处方配比为聚氧乙烯氢化蓖麻油(RH40)∶聚乙二醇-400(PEG400)∶肉豆蔻酸异丙酯(IPM)∶水=18∶6∶12∶64,维生素E质量浓度为0.004 mg·mL^-1,粒径为81.173 nm。结论:所建立的含量测定方法操作简便、准确;所制沙棘籽油微乳载药量较高,粒径较小,可显著增加沙棘籽油的溶解度,所得微乳稳定性好。
Objective:To establish a method for the determination of vitamin E in sea buckthorn seed oil microemulsion and optimize the formulation and ratio of sea buckthorn seed oil microemulsion.Methods:HPLC method was used to determine the solubility of sea buckthorn seed oil in different oil phases,emulsifiers and co-emulsifiers.The amount of water added was measured as the appearance of the system to become clear after dripping water phase,and the pseudo-ternary phase diagram was drawn,and the optimal formula composition was obtained.With the percentage of emulsifier,oil phase and co-emulsifier as independent variables,vitamin E content and particle size of sea buckthorn seed oil microemulsion were the response values.Box-Behnken response surface method was used to optimize the optimal formula ratio of sea buckthorn seed oil microemulsion.Results:The optimal formulation and ratio of sea buckthorn seed oil microemulsion was polyoxyethylene hydrogenated castor oil∶polyethylene glycol-400∶isopropyl myristic acid∶water=18∶6∶12∶64,the vitamin E content was 0.004 mg·mL^-1,the particle size was 81.173 nm.Conclusion:The method is simple and accurate for the determination of microemulsion of sea buckthorn seed oil.The prepared sea buckthorn seed oil has high drug loading and small particle size,which can significantly increase the solubility of sea buckthorn seed oil,and the obtained microemulsion has good stability.
