摘要
目的研究青果多糖组分CFW对胃癌细胞增殖、迁移、侵袭及凋亡的影响,并探讨其作用机制。方法提取青果水部位多糖组分CFW;将CFW(0.5、1.0、1.5 g/ml)处理胃癌细胞MKN4548 h,确定IC50;将ERBB信号通路抑制剂PF299804、DMSO分别与CFW共处理MKN45细胞24 h,标记为CFW+PF299804组、CFW+DMSO组;MTT法检测细胞的抑制率、增殖;Transwell小室检测细胞的迁移和侵袭;流式细胞术检测细胞的凋亡;Western blot检测细胞中P21、Bax、Twist、MMP-2、Bcl-2、ERBB的蛋白表达。结果与control组相比,CFW(0.5、1.0、1.5 g/ml)可呈浓度依赖性提高MKN45细胞的抑制率,选出IC50值为1.06 g/ml,且可明显抑制MKN45细胞的增殖、迁移和侵袭,促进凋亡,并上调P21、Bax,下调Twist、MMP-2、Bcl-2、ERBB;抑制ERBB信号通路可增强CFW对MKN45细胞的增殖、迁移和侵袭抑制和凋亡促进作用。结论青果多糖组分CFW可抑制胃癌细胞的增殖、迁移和侵袭,促进凋亡,其机制可能与抑制ERBB信号通路相关,将可为胃癌的治疗提供新的候选药物。
Objective To investigate the effects of green fruit polysaccharide constituents-CFW on the proliferation,migration,invasion and apoptosis of gastric cancer cells via ERBB signal pathway,and to explore its action mechanism.Methods The polysaccharide constituents-CFW extracted from the water content of the green fruit(0.5,1.0,1.5g/ml)was used to treat the gastric cancer cells-MKN45 for 48h to determine IC50.The ERBB signaling pathway inhibitors PF299804 and DMSO were used,together with CFW,to treat MKN45 cells for 24h,respectively,which were labeled as CFW+PF299804 group and CFW+DMSO group.MTT assay was used to detect the inhibitory rate and apoptosis of the cells and Transwell chamber was used to detect cell migration and invasion.Moreover,Flow cytometry was used to detect cell apoptosis and Western blot was used to detect the protein expression of P21,Bax,Twist,MMP2,Bcl2 and ERBB.Results As compared with control group,CFW(0.5,1.0,1.5g/ml)could increase the inhibition rate of MKN45 cells in a concentration-dependent manner.The IC50 value was 1.06g/ml,which could significantly inhibit the proliferation,migration and invasion of MKN45 cells,up-regulate P21 and Bax,down-regulate Twist,MMP2,Bcl2 and ERBB.The inhibition of ERBB signaling pathway could enhance the promotion effects on the proliferation,migration,invasion inhibition and apoptosis of CFW on MKN45 cells.Conclusion The green fruit polysaccharide constituents-CFW can inhibit the proliferation,migration and invasion of gastric cancer cells and promote apoptosis,and its action mechanism may be related to the inhibition of ERBB signaling pathway,which can be regarded as the candidate drug in treatment of gastric cancer.
