摘要
以咖啡因做内标物,建立了葡萄酒中柠檬黄、苋菜红、胭脂红、日落黄高效液相色谱内标法测定配制酒中4种人工合成色素的方法。样品经聚酰胺粉提取后,采用C18Nova-PakC18(3.9mm×15mm×5μm)色谱柱分离,以甲醇-0.02mol/L乙酸铵溶液为流动相进行梯度洗脱,流速为lmL/min,检测波长为470nm,柱温为30℃,在此条件下进行检测。结果表明,在0.05~0.1mg/kg的添加水平,柠檬黄、苋菜红、胭脂红、日落黄的加标回收率分别为99.5%~99.8%,99.2%~99.6%,94.8%~100.1%,92.1%~95.4%。相对标准偏差(RSD)分别为0.69%、0.52%%、0.66%、0.59%。所测配制酒中柠檬黄、苋菜红、胭脂红、日落黄的含量分别为1.27mg/g、1.33mg/g、1.80mg/g、1.47mg/g。采用该方法回收率高、准确度高、操作简便,可用于配制酒中柠檬黄、苋菜红、胭脂红、日落黄含量分析。
Coffee was used as the internal standard substance,a method of detecting of four kinds of synthetic pigment in wine including tartrazine,amaranth,carmine,sunset yellow by HPLC internal standard method was developed.After polyamide powder extraction,C18Nova-Pak C18(3.9mm×15mm×5(including m)chromatographic column was used in separation,0.02 mol/L ammonium acetate methanol solution was used as mobile phase in gradient elution,velocity was lmL/min,detection wavelength was 470 nm,column temperature was 30℃.Under the above conditions,at 0.05~0.1mg/kg range,the standard addition recoveries for tartrazine,amaranth,carmine,sunset yellow were 99.5%~99.8%,99.2%~99.6%,94.8%~100.1%,92.1%~95.4%,respectively.Relative standard deviation(RSD)were 0.69%,0.66%,0.59%,0.52%%.The content of tartrazine,amaranth,carmine,sunset yellow were 1.27mg/g,1.33mg/g and 1.80mg/g and 1.47mg/g.This method has high recovery rate,high accuracy and easy operation.It can be used to detect tartrazine,amaranth,carmine,sunset yellow content in wine.
