基于网络药理学当归黄柏治疗慢性前列腺炎的作用机制研究 被引量:20

摘要 目的:基于网络药理学方法探讨当归-黄柏治疗慢性前列腺炎(chronic prostatitis,CP)的作用机制。方法:应用中药系统药理学分析平台(TCMSP)并结合文献检索,筛选当归-黄柏中的活性成分与作用靶点,利用Uniprot数据库对靶点进行校正,应用GeneCards、OMIM数据库筛选疾病CP靶点,应用R语言软件筛选药物与疾病共同靶点,应用Cytoscape3.5.1软件构建药物-活性成分-靶点-疾病可视化网络图,应用STRING平台构建共同靶点蛋白互作(protein-protein interaction,PPI)网络,利用R语言软件通过基因功能注释(gene ontology,GO)和京都基因和基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)对核心靶标进行基因功能和通路的注释分析。结果:筛选出46个当归-黄柏的活性成分,预测相关靶点212个,其中与疾病有关的靶点159个,这些靶点主要涉及异源生物刺激反应、氧化与抗氧化反应等生物学过程,并主要富集在PI3K-Akt、丝裂原活化蛋白激酶(mitogenactivated protein kinase,MAPK)、低氧诱导因子-1(hypoxia inducible factor-1,HIF-1)等相关信号通路上。结论:通过网络药理学证实了当归-黄柏多成分、多靶点、多途径的作用特点,预测当归-黄柏治疗CP的可能作用机制,为进一步实验验证其作用机制提供了理论依据。 Objective:To explore the mechanism of Angelicae Sinensis-Phellodendri Chinrnsis in the treatment of chronic prostatitis(CP)based on the method of network pharmacology.Methods:The active components and targets in Angelicae Sinensis-Phellodendri Chinrnsis were screened by using the Systematic Pharmacological Analysis Platform for Traditional Chinese Medicine(TCMSP)combined with literature search.Targets were corrected by using the Uniprot database.GeneCards and OMIM databases were applied to screen the targets of disease.The common targets of drugs and diseases were screened by R software.The visual network diagram of drug-active component-disease target was constructed by Cytoscape3.5.1 software.The PPI network was constructed based on STRING platform.Annotation analysis of gene functions and pathways was performed on core targets by GO and KEGG by using R software.Results:46 active components of Angelicae Sinensis-Phellodendri Chinrnsis were screened and 212 related targets were predicted,of which 159 were related to diseases.These targets were mainly involved in biological processes such as heterologous biological stimulation response,oxidative and antioxidant response,and the targets were mainly enriched in PI3K-Akt,MAPK,HIF-1 and other related signaling pathways.Conclusion:The characteristics of multi-component,multi-target and multi-pathway of Angelicae Sinensis-Phellodendri Chinrnsis were confirmed by network pharmacology,and the possible mechanism of Angelicae Sinensis-Phellodendri Chinrnsis in the treatment of CP was predicted,which provides a theoretical basis for the further study of its active components and mechanism.
