摘要
目的探讨核转录因子GATA6蛋白信号传导和转录激活因子4(STAT4)在结肠癌中的表达及与临床特征和预后的关系。方法选取本院2011年3月-2015年8月收治的156例结肠癌患者为研究对象,收集156份癌组织标本及癌旁组织标本,行GATA6与STAT4蛋白的检测。采用χ^(2)检验分析GATA6与STAT4蛋白表达与临床病理特征的关系,并用Cox回归分析探讨结肠癌患者预后影响因素。结果结肠癌组织中GATA6高表达率为80.13%,明显高于癌旁组织23.72%(P <0.05);STAT4蛋白低表达率为78.21%,明显高于癌旁组织25.00%(P <0.05);肿瘤低分化、有淋巴结转移、TNM Ⅲ期~Ⅳ期的结肠癌患者GATA6高表达率和STAT4低表达率较高(P <0.05);预后单因素分析显示:GATA6高表达、STAT4低表达、TNM分期Ⅲ/Ⅳ期、低分化、有淋巴结转移的结肠癌患者生存时间明显缩短,上述差异均有统计学意义(P <0.05);多因素Cox回归分析结果显示,GATA6高表达、STAT4低表达、TNM Ⅲ/Ⅳ期、低分化、有淋巴结转移均为影响结肠癌患者预后的独立危险因素(P <0.05)。结论 GATA6与STAT4蛋白在结肠癌组织中表达水平与肿瘤分化程度、淋巴结转移、TNM分期等相关,GATA6高表达、STAT4低表达均为结肠癌患者预后独立危险因素,二者可能参与了结肠癌的发生发展。
Objective To investigate GATA6,signal transducer and activator of transcription 4(STAT4) in colon cancer tissues and its correlation with clinicopathological features and prognosis. Methods A total of 156 colon cancer patients admitted to our hospital from March 2011 to August 2015 were selected,and then the cancer tissues(n = 156) and adjacent tissues(n = 156) were collected from the patients. The protein levels of GATA6 and STAT4 in the selected tissues were detected.χ^(2) test was used to analyze the relationship between the expression of GATA6,STAT4 and the clinicopathological characteristics of patients,and Cox regression analysis was used to explore the prognostic factors of patients with colon cancer. Results The high expression rate of GATA6 was 80. 13%,which was significantly higher than that of adjacent tissues(23. 72%)(P <0. 05);and the low expression rate of STAT4 protein was 78. 21% in colon cancer tissues,which was significantly higher than that of adjacent tissues 25. 00%(P < 0. 05). The high expression of GATA6 and low expression of STAT4 in patients with poorly differentiated tumors,lymph node metastasis and TNM stage Ⅲ-Ⅳ were higher(P < 0. 05). Univariate analysis showed that the survival time was significantly shorter among patients with poorly differentiated tumor,lymph node metastasis,TNM Ⅲ-Ⅳ and high expression of GATA6 and low expression of STAT4,with the differences statistically significant(P < 0. 05).The multivariate Cox proportional hazards regression model showed that high expression of GATA6,low expression of STAT4,TNM stage Ⅲ/Ⅳ,low differentiation,lymph node metastasis were independent risk factors for the prognosis of colon cancer patients(P < 0. 05). Conclusion The up-regulation of GATA6 and down-regulation of STAT4 in the colon cancer patients are related to the differentiation degree,lymph node metastasis and TNM stage,and high expression of GATA6 and low expression of STAT4 are independent risk factors for prognosis of colon cancer patients,indicating their roles in the occurrence and development of the disease.
