摘要
目的研究桔梗多糖对桔梗皂苷D在体肠吸收的影响。方法大鼠随机分为对照组及桔梗多糖低、中、高剂量组,建立在体肠灌注模型,考察桔梗多糖对桔梗皂苷D吸收速率常数(K_(a))、表观吸收系数(P_(app))、口服药物吸收分数(fa)的影响。灌注后,取下大鼠肠段,免疫荧光染色法检测Claudin⁃1、Occludin、ZO⁃1表达。结果与对照组比较,桔梗多糖组Ka、P_(app)、f_(a)值升高(P<0.05,P<0.01),Claudin⁃1、Occludin、ZO⁃1表达下调(P<0.05,P<0.01),并呈剂量依赖性。结论桔梗多糖可改善桔梗皂苷D在体肠吸收,其机制可能与下调紧密连接蛋白表达有关。
AIM To study the effects of Platycodon grandiflorum polysaccharides on in situ intestinal absorption of platycodin D.METHODS Rats were randomly assigned into control group and low⁃dose,medium⁃dose,high⁃dose P.grandiflorum polysaccharides groups,the in situ intestinal perfusion model was established,after which the effects of P.grandiflorum polysaccharides on absorption rate constant(K_(a)),apparent absorption coefficient(P_(app))and absorption fraction of oral medicine(f_(a))of platycodin D were investigated.After the perfusion,the rat intestinal segment was collected,and Claudin⁃1,Occludin and ZO⁃1 expressions were detected by immunofluorescence staining method.RESULTS Compared with the control group,the P.grandiflorum polysaccharides groups demonstrated increased Ka,Papp and f_(a) values(P<0.05,P<0.01),and down⁃regulated Claudin⁃1,Occludin and ZO⁃1 expressions(P<0.05,P<0.01)in a dose⁃dependent manner.CONCLUSION P.grandiflorum polysaccharides can improve the in situ intestinal absorption of platycodin D,whose mechanism may contribute to the down⁃regulation of tight junction protein expressions.
