摘要
目的:研究西黄丸含药血清对三阴性乳腺癌细胞株MDA-MB-231细胞增殖、凋亡、迁移以及对环氧化酶2(COX-2)转录的影响,以期探讨西黄丸对三阴性乳腺癌细胞的作用。方法:大鼠灌胃给药制备西黄丸含药血清,实验设置大鼠空白血清组和西黄丸含药血清组,以含药血清干预三阴性乳腺癌细胞株MDA-MB-231,MTT法检测细胞增殖,流式细胞术检测细胞凋亡,细胞划痕实验检测细胞迁移,RT-PCR法检测COX-2、TNF-αmRNA表达。结果:西黄丸含药血清可以抑制MDA-MB-231细胞增殖,可以促进MDA-MB-231细胞凋亡,并且西黄丸含药血清可以降低MDA-MB-231细胞划痕愈合率,可以降低在炎性环境下MDA-MB-231细胞中COX-2 mRNA表达(P<0.05)。结论:西黄丸含药血清可以有效地抑制MDA-MB-231细胞增殖,促进其细胞凋亡,可能抑制细胞迁移,并且可以减少炎性因子的转录。
Objective:To study the effects of Xihuang Pills containing serum on cell proliferation,apoptosis,migration and COX-2 transcription of triple-negative breast cancer cell line MDA-MB-231,to investigate the effect of Xihuang Pill on triplenegative breast cancer cells.Methods:Rat serum containing Xihuang Pills was prepared by intragastric administration.Rat serum group(Control)and Xihuang Pill drug-containing serum group(XH)were set up to intervene in triple negative breast cancer cell line MDA-MB-231 with drug-containing serum.Cell proliferation was detected by MTT assay,apoptosis was detected by flow cytometry,cell migration was detected by cell scratch assay,and COX-2 and TNF-αm RNA expression was detected by RT-PCR.Results:Xihuang Pill-containing serum can inhibit the proliferation of MDA-MB-231 cells and promote the apoptosis of MDAMB-231 cells,and the serum containing Xihuang Pill can reduce the scratch healing rate of MDA-MB-231 cells.The expression of COX-2 mRNA in MDA-MB-231 cells was decreased in an inflammatory environment(P<0.05).Conclusion:Xihuang Pillcontaining serum can effectively inhibit the proliferation of MDA-MB-231 cells,promote its apoptosis,inhibit cell migration,and reduce the transcription of inflammatory factors.
